摘 要:[目的]探讨不同浓度对苯二胺原料和含对苯二胺的染发剂经皮肤接触对实验动物淋巴细胞的免疫效应和皮肤致敏作用。[方法]取浓度为0.3%、1.0%、3.0%、6.0%的对苯二胺原料和含对苯二胺为0%、1.5%、3.0%、6.0%的染发剂涂抹于小鼠耳背,连续3d,d4小鼠腹腔注射BrdU,d5摘取小鼠耳部淋巴结,制取单细胞悬液并计算淋巴细胞数,采用ELISA技术测定BrdU标记指数;取含对苯二胺为0%、1.5%、3.0%、6.0%的染发剂进行豚鼠皮肤变态反应实验。[结果]与阴性(溶剂)对照组小鼠相比,对苯二胺原料和含对苯二胺的染发剂处理的小鼠淋巴结重量、淋巴细胞数和BrdU标记指数均有显著的增加,在3.0%、6.0%浓度时均判为致敏性阳性;豚鼠皮肤变态反应实验结果显示含对苯二胺的染发剂在0%、1.5%、3.0%、6.0%浓度时致敏率分别为0%、0%、5%、15%,3.0%、6.0%浓度时致敏强度为弱致敏和轻度致敏。[结论]对苯二胺经皮肤接触可引起小鼠淋巴细胞明显增殖,含3.0%、6.0%对苯二胺的染发剂具有潜在的皮肤致敏性。[Objective]To study the effects of paraphenylenediamine on lymphocytes proliferation in mice and skin sensitization potency.[Methods]Mice were treated with 0.3%,1.0%,3.0% and 6.0% paraphenylenediamine(PPD)and four hair dyes contained 0%,1.5%,3.0% and 6.0% PPD at the dorsum of each ear once daily for three consecutive days.At the forth day,the mice were injected intraperitoneally with BrdU.At the fifth day,they were sacrificed and the draining auricular nodes were excised and weighed.Single lymphocytes suspensions of lymph node cells were prepared and lymphocytes number were counted.Used an enzyme-linked immunosorbent assay(ELISA)to measure the level of BrdU incorporation.Skin sensitization test in guinea pigs were executed to four hair dyes contained 0%,1.5%,3.0% and 6.0% of PPD.[Results]Compared with the vehicle-treated control group,the node weight,lymphocytes number and BrdU label index of the mice treated with PPD or hair dyes contained various concentration PPD were significantly increased.3.0% and 6.0% of PPD and hair dyes contained 3.0% and 6.0% of PPD showed positive responses;Hair dyes contained 3.0% and 6.0% of PPD were classified as weak and mild sensitizers in skin sensitization test in guinea pig.[Conclusion]Lymphocyte proliferation can be caused by PPD in skin contact.Hair dyes contained 3.0% and 6.0% of PPD have skin sensitization potency.
