国产磁珠核酸自动提取系统对HBV-DNA检测前处理的研究

摘　　要：目的:比较国产磁珠法核酸自动提取系统和传统的沉淀离心提取法提取血清HBV DNA的效果,探讨其应用潜力。方法:以两种方法平行提取50份血清HBV DNA,荧光定量PCR检测其提取核酸的拷贝数及CT值,进行方法对比试验;配制成不同浓度的HBV应用血清,用国产自动提取系统提取HBV DNA,评价方法的灵敏度、重复性及线性范围;向已知浓度的HBV阳性血清中定量加入溶血及脂血的阴性血清,再同样提取HBV DNA,观察方法的抗干扰特性。结果:两种方法平行提取血清HBV DNA,方法对比试验显示无显著差别;国产自动提取系统的重复性良好,定量检测的灵敏度及线性范围与现行方法一致;抗干扰能力强,溶血及脂血等常见干扰物对HBV DNA提取无干扰。结论:国产磁珠法核酸自动提取系统对血清HBV DNA的提取效果良好,操作简单,自动化程度高,重复性好,适宜用于临床HBV DNA检测前处理的核酸提取。Objective：To compare the effect of the domestic equipment of automatic nucleic acids extraction system with magnetic beads for the detection and pre-treatment of HBV-DNA,and to discuss its potential applications.Methods：Compare the effect of the domestic equipment of automatic nucleic acids extraction system with magnetic beads to that of the conventional precipitation and centrifugal extraction method for the detection and pre-treatment of HBV DNA on the sera from 50 patients.Analyze the sensitivity,reproducibility,linear range and the detection stability against interfering substances of the extraction method with the domestic equipment of automatic system.Negative serum of hemoglobin and triglyceride were added into the HBV negative serum whose concentration has been known,then HBV DNA was extracted in the same way to observe the anti-interference feature.Results：Contrast experiments showed no significant difference between the two methods.The domestic equipment of automatic nucleic acids extraction system has good repeatability,the sensitivity and linear range of quantitative detection were consistent with the current method.The common interferences in serum such as hemoglobin and triglyceride have no significant effects on the HBV DNA extraction with the domestic equipment of automatic system.Conclusion：The domestic equipment of automatic nucleic acids extraction system with magnetic beads has sound effects with easy operation and better automaticity,which is suitable for the clinical detection and pre-treatment of HBV-DNA.
