摘 要:以活体喷雾法作对照,研究了不同离体接种方法、不同接种浓度以及不同苗龄对甘蓝抗病性鉴定的影响.结果表明:采用环叶打孔离体叶片滴接法在甘蓝4~6片真叶期时,将浓度为1.0×108 cfu/mL的细菌悬浮液接种在直径1.5cm甘蓝叶片上,放到铺有湿滤纸的培养皿中密封后在28℃培养箱中培养5d后进行鉴定,其鉴定结果与活体喷雾法鉴定结果一致,可用于抗源筛选、品种(系)抗性鉴定及在抗病突变体筛选等方面.In order to identify black rot resistance in cabbage rapidly,different inoculation methods,bacterium concentrations and seedling stages were compared with in vivo spray as the control.A good method was identified.Its procedure included the following steps: in vitro punching was made on the leaves of cabbage plants with 4-6 expanded leaves,and the leaves with a diameter of 1.5 cm were inoculated with a bacterium suspension with a concentration of 1.0×10^8 cfu/mL.Then the leaves were transferred to Petri plates containing wet filter paper and cultured at 28oC in an incubator.The colonies on the inoculated leaves were observed 5 days after inoculation.The identification result was consistent with that of in vivo spray.It is concluded therefore that this method can be used in resistance source selection,variety/line resistance identification and screening for resistant mutants.
