摘  要:目的探讨干扰素调节因子-5(IRF-5)rs2004640位点T等位基因与类风湿关节炎(RA)的临床表现的相关性。方法采用聚合酶链反应-连接酶检测反应(PCR-LDR)对136例RA患者进行IRF-5rs2004640位点的单核苷酸多态性(SNP)分型。记录患者的临床表现、X线分期,检测红细胞沉降率、类风湿因子水平。结果 X线分期为Ⅲ+Ⅳ期组RA患者的T等位基因阳性率显著高于Ⅰ+Ⅱ期组(P=0.018)。关节外表现<3个的RA患者的T等位基因阳性率显著多于关节外表现≥3个的RA患者(P<0.01)。结论 IRF-5 rs2004640 T等位基因与RA患者关节破坏程度(X线分期)和关节外表现有关。Objective To study the correlation of interferon regulatory factor 5(IRF-5) gene rs2004640 T Alleles with the clinical manifestation of rheumatoid arthritis (RA). Methods The single-nucleotide polymorphism of IRF-5 rs2004640 was genotyped by polymerase chain reaction-tigase detection reaction(POR-LDR) in 139 RA patients. The clinical manifestations, X-ray stages, ESR, and levels of rheumatoid factor were all observed. Results The T allele frequency of IRF-5 rs2004640 was significantly higher in RA patients in Ⅲ+Ⅳ stage of X- ray than in those in Ⅰ + Ⅱ stage of X-ray(P=0. 039). The T allele frequency of IRF-5 rs2004640 in RA patients who had less than three extraarticular manifestations was significantly higher than that in patients who had 3 or more extraarticuiar manifestations (P〈0.01). Conclusion IRF-5 gene rs2004640 T Alleles is associated with bone damage degree (X-ray stage) and extraarticular manifestations in RA.
