摘 要:目的:观察葱白提取物纳米乳口腔喷雾剂对急性心肌缺血兔心电图及心肌梗死(简称心梗)面积的影响。方法:日本大耳白兔56只,随机分为假手术组、空白模型组、硝酸甘油组、葱白提取物组、空白纳米组、葱白纳米乳组和葱白纳米乳原位凝胶组,每组8只。术后及时舌下给药,硝酸甘油组、葱白提取物组、空白纳米组、葱白纳米乳组和葱白纳米乳原位凝胶组给予相应的制剂5喷,假手术组和空白模型组给予同体积的生理盐水。记录给药前及用药后0、1、5、10、15、30、60、120和180min的心电图,实验结束后处死动物,计算心梗面积。结果:①与模型组比较,各治疗组舌下给药后均可缩小兔冠脉结扎所致心肌梗死范围(P<0.05),葱白提取物及葱白提取物纳米制剂组减小心肌梗死面积的程度与硝酸甘油相当(P>0.05),葱白提取物纳米乳组和其纳米乳原位凝胶组与葱白提取物组未见明显差异(P>0.05);②对心电图的影响:与模型组比较,舌下给药后不同时间点各治疗组舌下给药后均可以使ST段显著降低(P<0.05);给药后1~5min,葱白提取物纳米乳组与硝酸甘油组比较作用相当(P>0.05),优于葱白提取物组(P<0.05)。葱白纳米乳原位凝胶起效时间为10~30min,葱白提取物起效时间为5~10min,葱白纳米乳组起效时间为0~5min,葱白纳米乳优于葱白提取物和葱白纳米乳原位凝胶,与硝酸甘油起效时间相当。结论:葱白提取物纳米乳口腔喷雾剂对ST段和心肌梗死面积的作用优于葱白提取物和葱白纳米乳原位凝胶,与硝酸甘油起效时间相当。Objective: To observe the effect of Chinese green onion (CGO) extract nanoemulsion oral spray on e- lectrocardiogram (ECG) and area of myocardial infarction in rabbits with acute myocardial ischemia. Methods Fifty-six Japanese male white rabbits were randomly divided into sham operation group, blank model group, nitroglycerin group, CGO extract group, blank nanoemulsion group, CGO extract nanoemulsion group, CGO nanoemulsion in situ gel group (n = 8). Rabbits in nitroglycerin group, CGO extract group, blank nanoemulsion group, CGO extract nanoemulsion group and CGO nanoemulsion in situ gel group were given 5 spray of corresponding prep- arations sublingually after operation, while rabbits in sham operation group and blank model group were given the same volume of normal saline. ECG was recorded before medication and 0, 1, 5, 10, 15, 30, 60, 120 and 180 minutes after medication. Rabbits were killed at the end of the experiments for calculating the area of myocardial infarction. Results: (1) Compared with that of model group, the area of myocardial infarction induced by coronary artery ligation was reduced in all medication groups after sublingual administration of corresponding preparations (P 〈0. 05). The degree of reduction of myocardial infarction area in CGO extract group and CGO extract nanoemulsion group was the same as that in nitroglycerin group (P〉0.05), there were no significant differences among CGO extract nanoemulsion group and its nanoemulsion in situ gel group and CGO extraction group (P〉0. 05) ; (2) Effects on ECG: Compared with that of model group, ST segment were significantly lowered after sublingual medication at different time points in each treatment group (P〈0. 05) ; effect of CGO extract nanoemulsion group was the same as that of nitroglycerin group 1-5 minutes after medication (P〉0. 05), but superior to that of CGO extract group (P〈0. 05).
