摘  要:目的:探讨凝血酶诱导大鼠CBP高表达影响大鼠血管平滑肌细胞(VSMCs)增殖的作用机制。方法:采用10-3、10-2、0.1、1、10U/ml的凝血酶干预体外培养的VSMCs 30min。RT-PCR法、蛋白印记法分别检测rCBPmR-NA和蛋白质的表达变化。流式细胞技术检测细胞周期,评价细胞增殖能力。结果:凝血酶可在mRNA和蛋白质水平明显诱导CBP高表达(P<0.05);凝血酶可以诱导VSMCs增殖,G0/G1期细胞比例显著降低(P<0.05),增殖指数(PI)明显增大(P<0.05),并且G0/G1期细胞比例与CBP含量呈负相关。结论:凝血酶可能通过上调CBPmRNA和蛋白质含量刺激VSMCs增殖。Objective: To explore the effect of thrombin on vascular smooth muscle cells (VSMCs) proliferation by inducing high-expression of cAMP response element binding protein (CBP). Methods: 10-3 U/ml, 10-2 U/ml, 0.1 U/ml,1 U/ml,10 U/ml thrombin interfered VSMCs culture for 30 minutes in vitro. Changes of rCBP mRNA rotein were detected by the methods of RT-PCR and western blot. The cell cycle was detected by flow cytomeevaluation of VSMCs proliferation. Results.. Thrombin might significantly induce high expression of CBP in mRNA and protein with dose dependence (P〈0. 05), and it might induce proliferation of VSMCs. The proportion of the cells in G1/G1 phase decreased significantly (P〈〈0. 05), the proliferation index was obviously increased (P〈 0. 05), and the proportion of the cells in G0/G1 phase was negatively correlated with the content of CBP. Conclusions: Thrombin might stimulate VSMCs proliferation through up-regulating CBP mRNA and protein content.
