摘 要:目的:探讨白花蛇舌草提取物对鼻咽癌细胞株CNE1的细胞毒性及其机制。方法:体外培养鼻咽癌细胞株CNE1,分别用不同浓度的白花蛇舌草水溶性提取物、脂溶性提取物和多糖提取物作用24、48 h,通过CCK-8法检测细胞存活率。分别用不同质量浓度的脂溶性提取物作用24、48 h,用Annexin V-FITC凋亡检测试剂盒检测细胞凋亡的情况。结果:3种白花蛇舌草提取物对鼻咽癌细胞株CNE1均有增殖抑制的作用,在一定的浓度范围内,呈剂量-效应关系,并存在时间-效应关系。3种提取物中,脂溶性提取物对鼻咽癌细胞株CNE1毒性最大,水溶性提取物次之,组间两两比较差异有统计学意义(P<0.05)。脂溶性提取物可诱导鼻咽癌细胞株CNE1凋亡,存在剂量-效应关系和时间-效应关系。结论:白花蛇舌草对鼻咽癌细胞株CNE1有显著的增殖抑制作用,其中以亲脂类提取物的作用最为明显,作用机制可能与诱导细胞凋亡有关。Objective: To investigate the cytotoxicity and its mechanisms of the extracts of Herba hedyotis to nasopharyngeal carcinoma cell line CNE1. Method: CNE1 cells were treated with water soluble, lipid soluble or polysaccharide extracts of Herba hedyotis, respectively. The potency of these extracts to inhibit cell growth was detected by CCK-8 assay. The apoptotic rate was detected by annxin V-FITC apoptotic detection assay. Results: All of the three kinds of extracts of Herba hedyotis inhibited cell proliferation of CNE1 cells in dose and time dependent manner. Among them, the cytotoxity of the lipid soluble extract was the most significant, and then the water soluble one. There were significant differences between different groups (P 〈 0. 05 ). The lipid soluble extract could induce the apoptosis of CNE1 cells in dose and time dependent manner. Conclusion: The extracts of Herba hedyotis had cytotoxicity to nasopharyngeal carcinoma cell line CNE1. And the lipid soluble extract had the most significant cytotoxicity, which might involve the induction of the cell apoptosis.
