摘 要:目的筛选日本血吸虫童虫外分泌蛋白,对日本血吸虫感染进行早期诊断。方法连续收集日本血吸虫感染后不同时间的动物血清,采用生物信息学方法确定血吸虫童虫外分泌蛋白基因,构建重组基因表达质粒,纯化外分泌蛋白,对重组蛋白的免疫原性进行分析,研究其特异性和敏感性。结果日本血吸虫童虫外分泌蛋白基因被插入到真核表达载体pAPtag2中,初步筛选获得了外分泌蛋白分子基因;重组蛋白的特异性为90.0%,敏感性为53.3%。结论重组血吸虫童虫外分泌蛋白,对日本血吸虫早期感染具有诊断价值。Objective To screen ultra-secreted proteins of schistosomula of Schistosomajaponicum and undergo early diagnosis of schistosoma infection. Methods Animal sera of schistosoma infection were collected at different time successively, genes of ultra-secreted proteins of schistosomula were identified to construct completed plasmids, and ultra-secreted proteins were purified to analyze immunogenicity, sensitivity and specificity of antigens for schistosoma infection in early time. Re- suits The genes of ultra-secreted proteins were inserted into plasmids of pAPtag2, specificity and sensitivity of reconstruc- tion proteins were 90.0% and 53.3%, respectively. Conclusions The ultra-secreted reconstructed proteins demonstrates the early diagnostic evaluation for schistosoma infections.
