HPLC法同时测定复方利血平片中苯磺酸氨氯地平和缬沙坦含量

摘　　要：目的:建立复方利血平片中苯磺酸氨氯地平和缬沙坦含量的测定方法。方法:色谱柱:Agilent Eclipse Plus C18柱(5μm,4.6 mm×250 mm),进样量:20μL,柱温:室温;流动相:甲醇-0.2%磷酸(70∶30),流速:1.0 mL.min-1;检测波长:237 nm。结果:苯磺酸氨氯地平在0.017 34～0.173 4μg、缬沙坦在0.401 5～4.015μg范围内,对照品色谱峰面积与进样量呈良好的线性关系,相关系数r分别为0.999 94与0.999 99,平均回收率分别为99.2%(RSD=1.29%,n=6)与99.6%(RSD=0.71%,n=6)。结论:该法快速、简便、重现性好,可用于复方利血平片中苯磺酸氨氯地平和缬沙坦含量的测定。OBJECTIVE：To establish a method for determining the contents of amlodipine besylate and valsartan in compound reserpine.METHODS：The chromatographic column of Agilent Eclipse Plus C18（5 μm,4.6 mm×250 mm） was used with injection volume of 20 μL.The column temperature was maintained at room temperature.The mobile phase consisted of methanol-0.2% phosphoric acid（70∶30） at a flow rate of 1.0 mL·min-1.The detection wavelength was set at 237 nm.RESULTS：The chromatographic peak area of the reference standard and sample volume showed a good linear relationship in a range of 0.017 34-0.173 4 μg for amlodipine besylate and 0.401 5-4.015 μg for valsartan,with correlation coefficient（r） being 0.999 94 and 0.999 99,respectively and average recoveries being 99.2%（RSD=1.29%,n=6） and 99.6%（RSD=0.71%,n=6）,respectively.CONCLUSION：This HPLC determination method is fast,simple and reproducible and it can be used in the determination of amlodipine besylate and valsartan in compound reserpine.
