胎盘内质网应激相关蛋白Calnexin与子痫前期发病的关系

摘　　要：目的探讨胎盘组织中内质网应激相关蛋白-钙联蛋白Calnexin(CNX) mRNA及蛋白表达与子痫前期发病的关系。方法剖宫产孕妇77例分为正常足月妊娠36例(A组),轻度子痫前期患者24例(B组)和重度子痫前期患者17例(C组)。RT-PCR技术检测胎盘组织CNX mRNA的表达,蛋白印迹法检测胎盘组织中CNX蛋白表达。结果 B、C组CNX mRNA表达水平高于A组[(1.3291±0.05687)(、3.4232±0.1078)vs.(0.5382±0.0345)](P<0.01);B、C组CNX蛋白表达水平高于A组[(1.5678±0.2315)、(2.1123±0.5310)vs.(0.5678±0.1089)](P<0.01)。结论 CNX参与了子痫前期的病理生理过程;内质网应激可能是子痫前期发病机制之一。Objective To investigate the relationship between pathogenesis of preeclampsia（PE） and the expressions of ensoplasmic reticulum stress-related protein calnexin and its mRNA.Methods Of 77 pregnant women undergoing Ceasarian section,36 cases were normal pregnant women（group A）,24 cases were diagnosed with mild PE（group B） and 17 cases were with severe PE（group C）.The expressions of calnexin mRNA and protein in placenta were detected with RT-PCR and Western blot.Results The calnexin mRNA expression levels were higher in groups of B and C than those in group A [（1.3291±0.05687） and（3.4232±0.1078） vs.（0.5382±0.0345）]（P0.01）.So did the expression levels of calnexin protein [（1.5678± 0.2315） and（2.1123±0.5310） vs.（0.5678±0.1089）]（P0.01）.Conclusion Calnexin is involved in the pathogenesis of preeclampsia.Endoplasmic reticulum stress may be one of the mechanisms for PE development.
