摘  要:目的观察地塞米松对体外培养人羊膜上皮WISH细胞糖皮质激素受体(GR)-α表达的调节。方法将体外培养的WISH细胞分为不同浓度地塞米松(10-8、10-7、10-6、10-5、10-4mol/L)作用48h组,和单一浓度地塞米松作用不同时间组(终浓度10-6mol/L,培养24、487、2h)。两组均设空白对照组。采用RT-PCR方法检测各组细胞中GR-αmRNA的表达。结果在不同浓度地塞米松作用48h后,10-4mol/L组细胞GRα mRNA表达强于空白组,其余各组细胞GR-α mRNA表达均弱于与空白组(P<0.01)。在地塞米松作用不同时间组,各亚组细胞GR-α mRNA表达与空白组相比,均明显减弱(P<0.01)。结论低浓度地塞米松可下调WISH细胞中GR-αmRNA表达;高浓度地塞米松可上调GRα mRNA的表达。GR-α mRNA表达不随地塞米松作用时间的延长而下降。临床上常规剂量和疗程应用地塞米松促胎肺成熟时,不会导致促分娩发动作用。Objective To observe the effects of dexamethasone on glucocorticoid receptor-α(GR-α) expression in the human amniotic epithelial cells,WISH cells.Methods The WISH cells were cultured in vitro with different concentrations of dexamethasone(10-8,10-7,10-6,10-5,10-4mol/L) for 48 h(group A) and a single concentration of dexamethasone(10-6mol/L) for different times of 24,48,and 72 h respectively.The results were compared to the blank control groups.The GR-α mRNA expressions in WISH cells were detected with RT-PCR.Results The expression of GR-α mRNA of WISH cells treated with 10-4mol/L DEX was higher than that of control group,which was loweer in those treated with the other concentration groups.The expression of GR-α mRNA was siginificantly lower in the cells treated with different times than that in those of blank controls(P0.01).Conclusion The expression of GR-α mRNA is decreased when treated with low concentration of dexamethason(≤10-6mol/L),which is increased when treated with high concentration of it(≥10-5mol/L).The decrease of GR-α mRNA expression is not reduced as the treating time prolongs.The clinical dose of dexamethasone for promoting lung paturity of the fetus could not lead to the onset of delivery.
