环孢菌素对大鼠牙龈结缔组织胶原及TIMP-1蛋白表达影响的实验研究

摘　　要：目的:观察环孢菌素(Cyclosporine,CsA)对大鼠牙龈结缔组织胶原和胶原酶TIMP-1表达的影响,探讨CsA对牙龈结缔组织胶原代谢的影响机制。方法:SPF级7周龄雄性Wistar大鼠80只,随机分为实验组和对照组,每组又分为10、20、30、40天亚组,实验组胃饲含CsA鲜牛奶,对照组胃饲等量鲜牛奶。经心灌注4%多聚甲醛内固定取材,制作下颌第一磨牙颊舌向石蜡切片,Masson三色改良法染色胶原,免疫组化PV两步法检测TIMP-1蛋白的表达。结果:实验组大鼠牙龈结缔组织内胶原纤维数量增多、直径增粗;牙龈结缔组织内TIMP-1蛋白表达上调。结论:环孢菌素导致牙龈结缔组织增生可能与TIMP-1蛋白表达上调、胶原堆积有关。Objective To observe the effect of Cyclosporine （CsA） on collagen and expression of the collagenase TIMP- 1 in gingival connective tissue of rats. To approach the collagen metabolic mechanism of CsA-induced gingival connective tissue overgrowth. Methods Eighty SPF grade male 7 weeks old Wistar rats were randomly divided into experimental group and control group medially, and each group was divided into 4 subgroups according to the duration of treatment （10,20,30 and 40 days）. The experimental objects were given intragastric administration of fresh milk including CsA,and the control ones were given intragastric administration of fresh milk the same as the experimental objects. After giving perfusion 4% paraform trans-heart to internal fixation, the specimens were get and made buccolingual paraffin sections at lower first molar. Collagen was detected using Masson trichrism improvement, and the expression of TIMP-1 using immunohistochemisty of PV. Results Cotlagenic number of the experimental group were significantly increased,and the collagenic diameter were significantly thickening. The expression of TIMP-1 of the experimental group were significantly up-regulated. Conclusion CsA-induced gingival connective tissue overgrowth is likely to associated with the up-regulated expression of the collagenase TIMP-1 and accumulating of collagen.
