摘 要:目的制备同时具有抗肿瘤药物缓释特性及修复骨缺损能力的抗肿瘤活性生物衍生骨,并通过体外实验研究该材料的缓释特性。方法取猪肱骨近端松质骨,经处理制成去抗原的生物衍生骨支架材料,再与顺铂明胶溶液复合,制成抗肿瘤活性的生物衍生骨材料;将该材料进行体外缓释实验,测定缓释液中的顺铂浓度并进行相应的肿瘤细胞抑制实验。结果去抗原的生物衍生骨支架材料呈白色,具有连续的多孔结构,具有天然骨的三维结构;复合明胶、顺铂后该材料呈淡黄色,孔隙为明胶、顺铂所填满;从材料的体外缓释曲线中可以观察到,顺铂在第1~3天释放药量较多,而后维持恒定的释放量,在2周内其缓释液对SHZ-88大鼠乳腺癌细胞的抑制率维持在50%以上,顺铂在局部的浓度可达100 g/ml左右,远高于一般腹腔给药所能达到的血药浓度。结论生物衍生骨材料是理想的药物缓释载体材料和骨修复的支架材料。将生物衍生骨材料与含有顺铂的明胶溶液复合制成的抗肿瘤活性生物衍生骨材料具有较好的缓释特性,能在体外抑制肿瘤细胞增殖。Objective To manufacture the anti-tumor bio-derived bone materials which have the characters of slow release anti-tumor drug and repair the bone defect after surgery.To study the characters of slow release anti-tumor drug in vitro experimental.Methods Sticks of pig cancellous bone were prepared into deantigenicity bio-derived bone materials by defatting,and deproteinisation,then composed with gelatin and CDDP.We used this material to observe the character of the control release in vitro,and measured the concentration of CDDP in the leachate and tumor inhibitory rate.Results Deantigenicity bio-derived bone material appeared white and porous,which had the structure of the nature cancellous bone.Anti-tumor bio-derived bone material appeared yellow,and all of the pores were filled with the compound of gelatin and CDDP.In vitro release experiment,CDDP from anti-tumor bio-derived bone materials appeared bursing release at 1-3 day,and maintain stable release in following 10 days,and the tumor inhibitory rate was higher than 50% in two weeks.The local concentration of CDDP was about 100 μg/ml,and higher than the blood drug level of intraperitoneal injection.Conclusion Deantigenicity bio-derived bone material was a perfect scaffold material to load the control drug release system and repair bone defect.Anti-tumor bio-derived bone material has good character of control release,and can inhibit the tumor cell proliferation in vitro.
