摘　　要：目的:观察槟榔碱对上皮细胞Hacat凋亡的影响。方法:用0,25,50,75,100,125μg/mL槟榔碱刺激上皮细胞Hacat24h,抽提DNA进行琼脂糖凝胶电泳和流式细胞术检测。结果:100和125μg/mL槟榔碱刺激后上皮细胞Hacat DNA梯状条带明显;50,75,100,125μg/mL槟榔碱诱导上皮细胞Hacat凋亡并呈剂量依赖性,随浓度升高进入晚期凋亡细胞的比例上升。结论:槟榔碱能通过诱导上皮细胞凋亡参与口腔黏膜下纤维性变的发生。Objective To observe the effect of arecoline on the apoptosis of Hacat cells.Methods Hacat cells were incubated with different concentrations of arecoline（0,25,50,75,100 or 125 μg/mL） for 24 h.DNA fragments and apoptosis were examined by agarose gel electrophoresis and flow cytometry,respectively.Results DNA fragments were obviously observed in groups treated with arecoline in a concentration of 100 or 125 μg/mL.The results of flow cytometry showed that arecoline induced apoptosis in a dosedependent manner.Conclusion Arecoline is able to induce Hacat cell apoptosis,which may contribute to the development of oral submucous fibrosis.
