漳州地区战备消毒包内细菌来源的实验研究

摘　　要：目的探讨沿海地区自然环境中战备消毒包内细菌来源。方法根据战备消毒包内腊样芽孢杆菌、人葡萄球菌、藤黄微球菌、沃氏葡萄球菌筛选战备仓库空气中同种细菌不同菌株,提取基因组DNA,以其为模板,对16SrRNA和23SrRNA的转录间隔区(Internal transcribled spacer,ITS)序列进行PCR扩增,将扩增产物回收测序,结果用Blast进行比对。结果腊样芽孢杆菌、人葡萄球菌、藤黄微球菌、沃氏葡萄球菌DNA的PCR扩增产物分别在850bp、1 000bp、1 100bp、1 000bp左右显现单一特异性条带;战备消毒包与空气同种细菌不同菌株之间的ITS序列相似性约为98%～99%。结论漳州地区自然环境中战备消毒包内细菌来源于空气细菌。Objective To investigate the origin of bacteria of sterile packages for war readiness under natural envi- ronment in Zhangzhou. Method Genomic DNAs of Bacillus cereus, Staphylococcus hominis, Micrococcus luteus, Staphylococcus warneri in sterile packages from war readiness and from air bacteria strains in war readiness ware- house were extracted, and then used as templates to amplify the 16 SrRNA and 23 SrRNA internal transcribed spac- er （ITS）sequences by PCR. The PCR products were extracted and sequenced, and the sequences homology analysis was performed with BLAST. Result Amplification products of DNA PCR from Bacillus cereus, Staphylococcus hom- inis, Micrococcus luteus, Staphylococcus warneri were about 850 bp, 1 000 bp,1 100 bp,1 000 bp respectively; The 16 SrRNA-23 SrRNA ITS sequences of bacteria strains in sterile packages shared 98% to 99% homology with those from the air bacteria strains in war readiness warehouse. Conclusion The bacteria in sterile packages for war readiness come from the air in war readiness warehouse under natural environment in Zhangzhou.
