摘 要:目的:研究山楂叶总黄酮(HLF)对大鼠脑缺血再灌注损伤的保护作用。方法:采用改良Zea Longa’s法复制大脑中动脉闭塞(middle cerebral artery occulusion,MCAO)模型。将健康雄性SD大鼠随机分成假手术、模型、尼莫地平和HLF高、中、低剂量组。用神经功能评分评价大鼠脑缺血再灌注后行为障碍;用2,3,5-三苯基氯化四氮唑(TTC)染色法测定梗死灶体积;用相应试剂盒测定脑组织内丙二醛(MDA)、一氧化氮(NO)含量及超氧化物歧化酶(SOD)活力。结果:与假手术组比较,模型组缺血脑组织内的MDA、NO含量显著升高,SOD活力显著降低(P<0.01);与模型组比较,HLF高、中剂量组脑梗死体积显著减少,行为学评分显著减少(P<0.01或P<0.05),HLF各剂量组能明显降低缺血脑组织内MDA、NO含量,增强SOD活力(P<0.01或P<0.05)。结论:HLF可能通过清除氧自由基、抗膜脂质过氧化和抗NO毒性作用起到抗脑缺血再灌注损伤作用。OBJECTIVE: To investigate the effects of total flavonoids of Crataegi Folium on cerebral ischemia-reperfusion injury in rats. METHODS: The model of middle cerebral artery occlusion (MCAO) was prepared by modified Zea Longa' s. Health male SD rats were randomly divided into sham operation group, model group, nimodipine group, total flavonoids of Crataegi Foli- um low-dose, middle-dose and high-dose groups. The behavioral disturbance was evaluated according to deficit scores; the infarction volumes were evaluated by 2,3,5-triphenyltetrzolium chloride (TTC) staining, the level of MDA, SOD, NO in the brain tissue of cerebral ischemia-reperfusion injury in rats were measured by the corresponding kits. RESULTS: Compared with sham operation group, the contents of MDA and NO in brain tissue in model group increased significantly, and the activity of SOD decreased significantly (P〈0.01). Compared with model group, the infarction volumes and etholodical score of HLF high-dose and middle-dose groups were decreased(P〈0.01 or P〈0.05). The content of MDA and NO in total flavonoids of Crataegi Folium groups decreased significantly, and the activity of SOD increased significantly(P〈0.01 or P〈0.05). CONCLUSION: Total flavonoids of Crataegi Folium may be enhance the ability of free radicals scavenging, anti-lipid peroxidation and anti-NO toxic injury, and then protects cerebral ischemic-reperfusion injury.
