摘 要:目的对高疟区"三热"疑似疟疾病人双重PCR检测与传统镜检法检测的比较,探讨双重PCR应用于临床诊断疟疾的前景。方法在高疟区门诊采集就诊的"三热"疑似疟疾病人末梢滤纸血,用chelex-100提取DNA模板进行双重PCR检测,同时与显微镜镜检厚血膜查找疟原虫进行比较。结果在334例疑似疟疾病例的血样中,双重PCR检测疟原虫敏感性为96.1%、漏检率为3.9%。镜检法检测疟原虫敏感性为86.1%、漏检率为13.9%。结论双重PCR在诊断疟疾上比传统镜检法敏感性高,适合于临床实验室对疟疾诊断。Objective To assess the value of double PCR in clinical diagnosis of suspected "three-fever "malaria cases by comparison with conventional microscopy in malaria endemic areas.Mthods The peripheral blood samples of 334 suspected "three fever" malaria outpatients were collected and chelex-100 DNA template were made and detected with double PCR and at the same time compared with microscopy of thick smear for malaria parasites.Results The sensitivity of double PCR in diagnosios of malaria was 96.1%,and a missing examination rate of 3.9%.While the sensitivity of conventional microscopy was 86.1% with a missing examination rate of 13.9%.Conclusion The sensitivity of double PCR in diagnosis of malaria is superior to conventional microscopy and suitable for clinical and labaroatory application.
