摘 要:目的对复方丹参方进行拆方、分离、纯化及配伍,观察复方丹参方不同组分及其配伍对2型糖尿病胰岛素抵抗(IR)的干预作用,并初步探讨其作用机制。方法以高糖高脂饲料喂养加小剂量链脲佐菌素腹腔注射法建立2型糖尿病IR动物模型,以复方丹参方不同组分及其配伍干预6周,于末次给药后分别检测各组大鼠空腹血糖、血脂、空腹胰岛素及氧化应激水平。结果丹参酚酸、三七皂苷及其配伍组分丹参酚酸+三七皂苷和丹参酚酸+三七皂苷+冰片,能够明显调节糖脂代谢,降低游离脂肪酸(FFA)表达,增强机体抗氧化能力,提高胰岛素敏感性;而丹参酮作用不明显。结论丹参酚酸、三七皂苷是复方丹参方中改善IR的主要活性部位,其作用与调节脂代谢、抑制FFA分泌、抗氧化应激等有关。Objective To investigate the different fractions and compatibilities of Fufang Danseng formulas on insulin resistance(IR) in type 2 diabetic rats.Methods Diabetic rats with IR were established by intraperitoneal injection with 30 mg/kg streptozotocin and a high-carbohydrate/high-fat diet.After 6 weeks treatment,the fasting blood glucose(FBG),insulin level,total cholesterol(TC),triglyceride(TG),low density lipoprotein-cholesterol(LDL-C),high density lipoprotein-cholesterol(HDL-C),free fatty acid(FFA),malondialdehyde(MDA),and superoxide dismutase(SOD) in serum were assayed.Insulin sensitivity index(ISI) was also calculated.Results Treatment with salvianolic acids(SAL),panax notoginseng saponins(NOT),SAL+NOT(SN),or SAL+NOT+borneol(SNB) significantly decreased the levels of FBG,insulin,TC,TG,LDL-C,MDA,but effectively increased the levels of ISI,HDL-C,SOD in diabetic rats.Conclusion Both of SAL and NOT can exert the beneficial effects on hyperglymia and insulin resistance through regulation of lipids metabolism,inhibition of FFA secretion,as well as anti-oxidation.
