益赛普对大鼠心肌缺血再灌注损伤中炎症相关细胞因子含量的影响

摘　　要：目的:探讨益赛普(etanercept)对结扎冠脉左前降支所致大鼠急性心肌缺血再灌注损伤的保护作用以及对炎症细胞因子的影响。方法:结扎大鼠冠状动脉左前降支30min,再灌注120min复制大鼠心肌缺血再灌注损伤模型,取心肌组织,HE染色观察大鼠心肌组织形态的变化,酶联免疫吸附法检测心肌组织中的肿瘤坏死因子-α(tumor necrosis factor-α),白细胞介素-10(interleukin-1,IL-10)与白细胞介素-6(interleukin-6,IL-6),血清中的肌钙蛋白-I(cardial intropin-I)的含量。结果:与模型组相比,益赛普组的心肌炎症反应较轻,心肌中肿瘤坏死因子,血清中IL-1,IL-6和肌钙蛋白的含量较低,差别有统计学意义。结论:益赛普对缺血再灌注损伤的心肌细胞具有的保护作用是通过作用抗炎症反应。Objective： To investigate the effect of Etanercept on inflammatory cytokines in rats with ischemia-reperfusion injury.Methods： A total of 36 male Wistar rats were randomly divided into 3 groups： Sham group,I/R group,Etanerceptan group.Ligation of left anterior descending coronary was performed for 30 min and then the perfusion was carried out for 2 h to establish ischemia-reperfusion（I/R） injury model.Myocardium tissue was collected and stained by HE staining.Morphological change was observed.Expression levels of TNF-α,troponin,IL-1 and IL-6,IL-10 were detected by ELISA.Results： In Etanercept group the expression of troponin,IL-1 and IL-6,IL-10 were significant lower compared with those of I/R group（P0.05）.Conclusions： Etanercept can protect myocardial cells by anti-inflammation.
