摘  要:目的:观察大鼠在严重的急、慢性应激条件下对学习和记忆的影响,这种影响是否会伴有海马内的Nestin、NPY的同步减少。方法:实验以捆绑并倒悬复合刺激3d和21d建立大鼠急、慢性应激模型,Morris水迷宫检测模型大鼠空间学习和记忆的能力;放射免疫方法测定大鼠血浆肾素、血管紧张素Ⅱ水平;免疫组织化学染色观察比较各应激大鼠海马区域NPY、Nestin、NF200阳性细胞表达。结果:急、慢性应激对大鼠的学习记忆能力均明显减退,海马组织内细胞明显受到损害,并以急性应激组更严重,表现为CA3区的Nestin、NPY表达和阳性细胞比例明显减少,而慢性应激大鼠缺乏表达NF200的成熟神经元。结论:成体海马神经组织内的NPY和细胞骨架蛋白系统共同参与了应激早期学习记忆能力减退和持续性脑损害的过程。Objective: To investigate the expression change of neuropeptide Y(NPY) and Nestin in hippocam-pus under severely acute and chronic stress,and its relationship with learning and memory.Methods: Acute and chronic stress rat models were established by combined restrained and inverted stress at 3 and 21days.All rats were tested to observe the influence of stress on learning and memory by the Morris water maze test.Radioimmunoassay was used to analyze PRA and Ang Ⅱ levels of the plasma of all model rats.Immunohisto-chemistry and cellular morphology were applied to observe and analyze hippocampal tissue,cell morphology,neurogenesis and the distribution of Nestin,NF200,NPY expression in the DG and CA1~4 regions of the hippocampus of the rats.Results: The ability of learning and memory significantly decreased in acute and chronic stress rats.In addition,hippocampal cells were damaged,particularly in the acute stress rats,and the number of Nestin and NPY-positive cells in the CA3 region significantly decreased.Furthermore,mature NF-200-positive neurons were absent in the chronic stress rats.Conclusion: The NPY and cytoskeletal protein system equally contribute to stress-induced early learning and memory deficiency,as well as sustained cerebral injury in the adult hippocampus.
