摘 要:目的建立用于具有抗菌活性中草药筛选的、简单有效的抗菌药物敏感性试验方法。方法在微量反应板上采用药基(药物和半固体培养基)混合试验法以丁香、黄连、虎杖和大青叶等抗菌中草药的水提液及乙醇提取液对金黄色葡萄球菌标准菌株、淋病奈瑟菌和白色念珠菌质控菌株进行抗菌试验,同时应用传统的琼脂平板稀释法作为对照,且重复试验3次。结果测试的4种中草药针对金黄色葡萄球菌ATCC25923的抗菌活性在1:128~1;512稀释度,针对淋病奈瑟茼199622在1:2稀释度,针对白色念珠菌199713在1:64~1:512稀释度,与对照方法相比抗菌活性多为相同或有一个稀释度的差异,仅有两个结果为两个稀释度差异。结论微量半固体琼脂稀释法用于抗菌活性中草药的筛选具有操作简便、结果易于判读的优点。Objective To establish a simple and effective antimicrobial susceptibility test for screening Chinese herbal medicine with anti-microbial activity. Methods Semisolid agar micro-dilution method was established for evaluate the an- timicrobial activity of herbal medicine,which mixe^l the semisolid agar medium with herbal extract and bacterium suspen- sion to allow different concentration of herbal medicine being tested. The agar dilution method was used as control. Results The antimicrobial activity of the four herbal medicine tested was 1 : 128 to 1 t 512 against Staphylococcus aureus ATCC25923,1 , 2 against Neissria gonorrhoeae 199622 and 1 : 64 to 1:512 against Candida albicans 199713 respectively. The majority MIC values of the two methods were the same or one dilution difference,and only two results two dilution difference. Conclusion The semisolid agar micro-dilution method could be excellent substitute for screening anti-microbial activity of Chinese herbal medicine.
