摘  要:以龙眼体细胞胚胎发生8个不同发育阶段同步化胚性培养物为材料,采用荧光定量PCR方法,分析两个乙烯受体基因(DL-ETR1和DL-ERS1)的mRNA动态变化水平。试验结果表明:DL-ETR1表达量整体的变化趋势呈近似"W"状,松散型的胚性愈伤组织(Stage 1)的表达量最高,心形胚(Stage 6)阶段的表达量最低;龙眼体胚发生过程的8个阶段均有DL-ERS1基因的表达,子叶形胚(Stage 8)的表达量最高,其它7个阶段的表达量都很低,鱼雷形胚(Stage 7)的表达量极低,几乎不表达。DL-ETR1与DL-ERS1有着不同时期的表达模式,并且可能起到结合乙烯,调节龙眼体胚对乙烯敏感性的综合作用。Eight different synchronized embryogenic cultures of developmental stages during somatic embryogenesis were used as materials to analyze the dynamical mRNA transcription levels of two ethylene receptor genes(DL-ETR1 and DL-ERS1)by the Quantitative PCR Method in Dimocarpus longan.The results showed that the whole trend of DL-ETR1 expression was approximately a "W"-shaped curve;the peak mRNA transcription level of DL-ETR1 occurred at the friable-embryogenic callus stage,and the valley mRNA transcription level of DL-ETR1 occurred at the heart embryo stage;DL-ERS1 expressed at the eight different stages during somatic embryogenesis;the expression levels of seven stages were very low except the cotyledonary embryos stage;the peak mRNA transcription level of DL-ERS1 occurred at the cotyledonary embryos stage,and the lowest mRNA transcription level of DL-ERS1 occurred at the topedo embryos,almost without expression.The two ethylene receptor homolog genes had stage-specific expression patterns,and they probably played a comprehensive role in combining ethylene and regulating the sensitivity of longan somatic embryos to ethylene.
