摘  要:利用ELISA法检测了抗虫棉益农二号不同时期叶片中的Bt蛋白的含量,以及棉田中棉蚜和龟纹瓢虫体内Bt蛋白的含量;利用室内Bt棉叶片饲养试验,观测棉蚜和龟纹瓢虫的生命参数。结果表明:抗虫棉真叶中Bt毒蛋白的含量随棉株发育而增加:从三叶期的121ng/g增加到盛花期的304ng/g,但子叶的含量高于早期的真叶。相应地,在抗虫棉田中的棉蚜和龟纹瓢虫体内均检测到了Bt毒蛋白的存在。棉蚜体内最高检测到6ng/g的水平,平均含量2.8~3.7ng/g;而龟纹瓢虫中Bt的含量高于棉蚜:四龄幼虫平均为8.4ng/g,雌性成虫体和雄性成虫分别为16.9ng/g和27.8ng/g。Bt棉上的棉蚜若虫繁殖力和蜕皮率略高于常规棉,若虫生长历期缩短0.5d左右;用Bt棉田的棉蚜饲喂的龟纹瓢虫,其生长发育、存活率以及繁殖力等未受到影响。The expression of CrylAc or CrylAb toxin in different leaves' stages of transgenic Bt cotton (AgroSciences II) and Conventional cotton were detected by EILSA assay. The Bt toxic protein content in the body of Aphids and P. Japonica was measured. We observed the life parameters changement of the relative insects which was fed on different food source via the In- door feeding experiment. The results demonstrate that the toxic protein CrylAc/CrylAb's quantitative expression in Bt cotton's leaves increased with the Bt cotton growth:from the three leaf stage (121 ng/g) increased to bloom stage (304 ng/g),but the cotyledon leaf were higher than other organs. Accordingly, the Bt toxin in the body of Aphids and P. Japonica could be detected in the pest-resistant cotton. The highest Bt protein content of the Aphids is 6 ng/g,the average content is 2.8-3.7 ng/g. The contentration of the Aphids when it is at the fourth instar larvae and female and male adult P. japonica is 8.4,16.9,27.8 ng/g respectivly on aging in Bt cotton. The effective transmission of the Bt toxic protein is significant. In addition,indoor insect feeding test indicated that the Aphids' nymphal period on Bt cotton was shorter 0. 5 d, the Reproduction and Molting rate was slightly higher compared with conventional cotton. However, when P. Japonica was fed on cotton aphis which collected from the transgenic Bt Cotton plots,we found that the Ladybirds' development and survival from hatching to emerge as well as the Reproductive ability was not significantly different from those of the Control.
