摘  要:【目的】优化兰州百合组织培养体系,提高其植株再生率,建立高频植株再生体系。【方法】采用正交试验设计,以鳞片为外植体,以MS和1/2MS培养基为基本培养基,附加不同浓度NAA(0.2~1.0mg/L)、6-BA(0.5~2.0mg/L)、KT(0.1~1.0mg/L)、IBA(0.2~1.0mg/L)、IAA(0.2~1.0mg/L),对兰州百合进行鳞茎诱导、增殖和生根培养,筛选最佳激素配比。【结果】6-BA浓度过低或过高均不利于小鳞茎形成,当1.0mg/L6-BA与0.2~1.0mg/LNAA配比时,鳞茎诱导率和平均生成鳞茎数较高;随着NAA浓度的增加,平均生成鳞茎数不断增加,其中以1.0mg/L6-BA+1.0mg/LNAA组合的平均小鳞茎最多,为5.9个。与0.5~1.0mg/LKT+0.2NAAmg/L组合相比,添加0.5~1.5mg/L6-BA+0.5~1.0mg/LKT或0.2NAAmg/L组合的培养基更利于芽增殖,但其丛芽长势较弱,其中以添加0.5mg/L6-BA+0.2mg/LNAA培养基的丛芽长势和小鳞茎长势较好,平均分化鳞茎较多。在以MS、1/2MS为基本培养基、分别添加0.2~1.0mg/LIAA或IBA的培养基中,随着IAA或IBA浓度的升高,鳞茎生根率不断降低,根质量不断下降,而平均根数和根长表现有所不同。从根的质量来看,MS培养基的根质量不如1/2MS培养基;而在1/2MS培养基中,1/2MS与IBA组合优于与IAA组合。【结论】鳞茎诱导的最佳培养基为MS+1.0mg/L6-BA+NAA0.5~1.0mg/L,不定芽增殖的最适培养基为MS+0.5mg/L6-BA+0.2mg/LNAA,最佳生根培养基为1/2MS+0.2mg/LIBA,生根效果最好,生根率为100.0%,根中等长度、粗壮,质量最好。[Objective]The present experiment was conducted to optimize tissue culture conditions for Lilium davidii var. Unicolor (Hoog) Cotton, to improve plantlet regeneration rate and to establish high efficient plant regeneration system from scales. [Method]Scales of L. davidii were used as explants for the induction, multiplication and rooting of bulbs in basic MS medium containing different combinations and concentrations of NAA(0.2-1.0 mg/L), 6-BA(0.5-2.0 mg/L), KT(0.1-1.0 mg/L), IBA(0.2-1.0 mg/L) and IAA(0.2-1.0 mg/L). [Result]The rate of induction and average number of induced bulbs was higher in the medium containing 1.0 mg/L 6-BA and 0.2-1.0 mg/L NAA. The average number of induced bulbs increased with increasing concentration of NAA in medium and reached the highest (5.9) in the medium containing 1.0 mg/L 6-BA+1.0 mg/L NAA. Compared to combination of 0.5-1.0 mg/L KT+0.2 NAA mg/L in medium, the combination of 0.5-1.5 mg/L 6-BA+0.5-1.0 mg/L KT or 0.2 mg/L NAA was more beneficial to the multiplication of bulbs, but the multiplied cluster buds of bulb showed weak growth, however, the cluster buds grew well in the medium containing 0.5 mg/L 6-BA+0.2 mg/L NAA. Supplementing 0.2-1.0 mg/L IAA or IBA in MS and 1/2MS medium, respectively, the rooting rate in bulbs and quality of root decreased with increase in IAA or IBA concentration, however, the average number of roots and root length showed different trends. The quality of root was better in 1/2MS medium supplemented with IBA than that in MS medium with IAA. [Conclusion]The MS medium containing 1.0 mg/L 6-BA + 0.5-1.0 mg/L NAA was the best for induction of bulbs, MS medium containing 0.5 mg/L 6-BA + 0.2 mg/ L NAA for bud multiplication and 1/ 2 MS+0.2 mg/L IBA for the rooting which resulted in fine growth of roots with 100.0% rooting rate.
