摘  要:目的研究人工麝香对金黄色葡萄球菌的生长代谢的影响,人工麝香质量生物活性评价新方法。方法采用微量热法,测定37℃时金黄色葡萄球菌在不同浓度的人工麝香作用下的热谱曲线,并获得其相应的生长速率常数(k)、最大发热功率(Pm)及其出现时间(tm)、总发热量(Qt)等热动力学参数,经主成分分析和回归分析等化学计量学方法综合评价人工麝香对金黄色葡萄球菌生长代谢的影响。结果人工麝香对金黄色葡萄球菌生长代谢具有明显的抑制作用,这种抑制作用主要受第一、二指数生长期最大发热功率出现时间(tm1、tm2)的影响。结论通过分析这两个主要参数的数值变化,能更方便、快捷和准确地评价人工麝香抑制金黄色葡萄球菌活性,为进一步评价人工麝香及其他药物的抗菌生物活性提供了有用的方法和基础。OBJECTIVE To investigate the effect of artificial musk on Staphylococcus aureus growth, and tn search new method for evaluating bioactivity of artificial musk. METHODS The power-time curves of S. aureus affected by different concentrations of artificial musk were recorded by microcalorimetry at 37℃. The thermokinetic parameters such as the growth rate constant (k), the maximum power output ( Pm) and its appearance time ( tm ), and the total heat output ( Q1) were obtained. Chemometric analyses such as principal component analysis (PCA) and regression analysis (RA) were employed to evaluate the effect of artificial musk on S. aureus growth. RESULTS Artificial musk had inhibitory action on S. aureus growth. The effect was mainly ifnluenced by the appearance time of the maximum power output in the first and the second exponential growth phases (t1m, t1m ). CONCLUSION The anti-S, aureus activity of artificial musk can be easily, quickly and exactly evaluated by analyzing the change of t2m, and t2m, This work provides useful method and foundation for further evaluating the antibacterial activity of artificial musk and other drugs.
