摘  要:目的应用代谢组学的方法,分析系统性红斑狼疮(SLE)患者血清的代谢物特征并构建疾病血清代谢物诊断模型。方法应用600MHzNMR检测64例SLE患者和35例健康志愿者血清样本中代谢物的差异,采集代谢物指纹图谱。并结合模式识别分析方法构建SLE诊断模型。结果 SLE主要影响脂类、氨基酸及能量代谢。PCA及PLS-DA均可以显著区分SLE与健康对照组,PLS-DA模型的预测率Q2=0.393。且O-PLS-DA模型对诊断具有很高的特异性(97.1%)及敏感性(60.9%)。结论代谢组学技术结合模式识别分析方法有望成为临床诊断SLE及病情监测的新方法。【Objectives】 To analyze serum metabolic profile of patients with SLE and to establish metabolic diagnosis model based on metabolomics approaches.【Methods】 600 HzNMR spectroscopy was applied to detect serum of 64 SLE patients and 35 health controls.Combined with multivariate pattern recognition analysis it can cluster the groups and establish disease-specific metabolic diagnosis model.【Results】 There is an alteration in lipid,amino acid and energy metabolism.PCA and PLS-DA model were capable of distinguishing SLE and healthy control and the Q of PLS-DA model is 0.930.And the OPLS-DA model was able to predict SLE diagnosis for SLE with a sensitivity and specificity of 60.9% and 97.1% respectively.【Conclusions】 Metabolomics approaches and multivariate pattern recognition analysis hold the potential to be a new tool to diagnosis and mornitoring SLE.
