摘 要:目的:观察异氟烷对罗哌卡因、氯普鲁卡因致小鼠惊厥及惊厥后情感行为和学习记忆的影响。方法:40只小鼠随机平行进入罗哌卡因、氯普鲁卡因2个实验。每个实验又随机分为生理盐水组(NS组)、异氟烷组(Iso组)(n=10)。各组小鼠先腹腔注射(intraperitoneal injection,ip)95%致惊厥剂量的罗哌卡因(86 mg/kg)、氯普鲁卡因(250 mg/kg),出现惊厥后立即ip NS(0.8 ml/kg)或Iso(0.8 ml/kg),观察小鼠惊厥持续时间及惊厥后在旷场实验(open filed test,OFT)、避暗实验(passive avoidancetest,PAT)中的行为改变。结果:在罗哌卡因、氯普鲁卡因实验中,Iso组与NS组相比:惊厥持续时间明显缩短(P<0.05,P<0.01),死亡数无明显差异(P>0.05)。在OFT中,中央格停留时间、穿格次数、修饰次数、粪便粒数的差异均无统计学意义(P>0.05)。在PAT中,记忆潜伏期和错误次数均无明显改变(P>0.05)。结论:异氟烷能够减轻罗哌卡因、氯普鲁卡因所致的惊厥,但对惊厥后小鼠短期的情感行为和学习记忆无明显影响。Objective To observe the effects of Isoflurane on ropivacaine or chloroprocaine-induced convulsion and influences on emotion,behavior,learning and memory after convulsion.Method Total 40 mice were randomly divided into ropivacaine and chloroprocaine experiments,each of which was divided into normal saline group(NS group) and Isoflurane group(Iso group),10 in each group.Ropivacaine or chloroprocaine was administered intraperitoneally,when the convulsion happened,normal saline or isoflurane were administered intraperitoneally.Then to observe convulsion duration and the influences on emotion,behavior,learning and memory in open filed test(OFT) and passive avoidance test(PAT) after convulsion.Results The convulsion duration was shorten by Isoflurane.There was no significant difference in OFT and PAT between Iso group and NS group.Conclusion Isoflurane can antagonize the ropivacaine or chloroprocaine-induced convulsion and has no influences on emotion,behavior,learning and memory shortly after convulsion.
