摘 要:用PCR技术扩增藏绵羊线粒体DNA(mtDNA)控制区时,发现藏绵羊群体中存在个体间和个体内的mtDNA长度变异现象。DNA测序表明,长度变异发生在mtDNA D-loop区靠近tRNApro220~520 bp的位置,由该区域75 bp的串联重复序列的重复数目的差异造成。在检测的27个个体组成的藏绵羊群体中,四个串联重复的个体有23个,占总群体数的85.2%;三个串联重复的个体有3个,占11.1%;既有三串联重复又有四串联重复的异质性个体有1个,占3.7%。线粒体DNA异质性现象产生的原因和意义有待进一步研究。It was detected that there existed variations about the mtDNA length among the individuals of Tibetan sheep or in certain individuals when their mitochondrial DNA were amplified using PCR.DNA sequence analysis revealed that the inter-and intra-individual length variation occurred at D-loop near tRNApro 220-520bp,and it was caused by the different number of tandem repeats with 75bp in that region.In this detection of 27 Tibetan sheep,23(85.2%) individuals had four tandem repeats,3(11.1%) individuals had three tandem repeats and 1(3.7%) individual had both three and four tantem repeats.More research is needed to clarify the reason and effect of the heteroplasmy about mtDNA.
