益肾填精助孕方药对肾虚胚胎着床障碍小鼠子宫内膜MMP-9及TIMP-3蛋白表达的影响

摘　　要：目的:观察益肾填精助孕方药仙子益真胶囊对肾虚胚胎着床障碍模型小鼠子宫内膜腺上皮细胞、腔上皮细胞和基质细胞MMP-9及TIMP-3的蛋白表达影响,探讨其可能作用机制。方法:建立肾虚胚胎着床障碍小鼠模型,分别予仙子益真胶囊低剂量、中剂量、高剂量、戊酸雌二醇片及生理盐水灌胃,免疫组化法检测MMP-9和TIMP-3的蛋白表达。结果:MMP-9蛋白及TIMP-3蛋白在模型组与正常组的子宫内膜腺上皮细胞、腔上皮细胞中表达有显著性差异(P<0.05)。MMP-9蛋白及TIMP-3蛋白在模型组与仙子益真胶囊各组的子宫内膜腺上皮细胞、腔上皮细胞中表达有显著性差异(P<0.05)。MMP-9蛋白及TIMP-3蛋白在仙子益真胶囊低、中剂量组小鼠子宫内膜腺上皮、腔上皮表达与戊酸雌二醇片组比较无显著性差异。TIMP-3蛋白在仙子益真高剂量组小鼠子宫内膜腺上皮、腔上皮表达与戊酸雌二醇片组比较有显著性差异(P<0.05)。结论:益肾填精助孕方药可以改善着床障碍小鼠子宫内膜MMP-9、TIMP-3的表达,从而调节滋养细胞对子宫内膜的侵入程度,促进胚胎着床。Objective： To observe the effect of kidney-tonifying and pregnant-promoting traditional Chinese herbs Xianziyizhen capsules on the express of endometrium protein of MMP-9 and TIMP-3 of luminal epithelium and stromal cells of mice which were modeled with dysfunction of embryo implantation. Methods： Establish mice model with dysfunction of embryoimplantation because of kidney-deficiency, and fed them with Chinese herbs Xianziyizhen of low dose, normal dose, high dose and Progynova, normal saline,respectively. Then use the detection of immunohis-tochemistry to detect expression of endometrium protein of MMP-9 and TIMP-3. Results： Expression of protein MMP-9 and TIMP-3 of epithelium cells and luminal epithelium cells shows significant differences between Chinese medicine group and the model group （P〈0.05）, and it shows no obvious differences between Chinese medicine group and Progynova group. Conclusion： Kidney-tonifying and pregnant-promoting traditional Chinese herbs can improve the express of endometrium protein of MMP-9 and TIMP-3 of mice which with dysfunction of embryo-implantation, thus to regulate trophoblast invasion degree of endometrial and to promote embryo implantation as the ultimate goal.
