摘 要:目的:观察葱白提取物对脂肪变性肝细胞模型中PPAR-α及PGC-1表达的影响,探讨其防治脂肪肝的作用机制。方法:建立游离脂肪酸诱导的细胞脂肪变性模型。用不同浓度葱白提取物进行干预24h后,MTT检测的细胞活性;油红O染色观察细胞内的脂滴;生化细胞内TG、ALT、AST含量;RT-PCR和Western Blotting分别检测细胞内PPAR-α、PGC-1基因的mRNA和蛋白表达。结果:游离脂肪酸诱导24h后,油红O染色显示脂肪变性;模型组TG含量较对照组显著升高,差异有统计学意义(P<0.01);各组转氨酶没有明显变化;同时胞内PPAR-α、PGC-1的mRNA、蛋白表达降低。而葱白提取物各剂量组胞内的TG较模型组显著降低,差异有统计学意(P<0.05,P<0.01),并且胞内PPAR-α、PGC-1的mRNA、蛋白表达显著升高,与模型组相比差异有统计学意义(P<0.05,P<0.01)。结论:葱白提取物对脂肪变性肝细胞有明显的保护作用,其机制可能与PPAR-α、PGC-1基因表达上调有关。Objective: To observe effects of the extract of onion on PPAR- α, PGC-1 expression in fatty degeneration of liver cell model, and to explore the possible molecular mechanism. Methods: With free fatty acid induced HepG-2, established steatosis model. With different concentrations of fresh onion extract for 24 hours, cell viability detected by MTT, rathonum red staining of intraceUular lipid droplets. Intracellular TG, ALT and AST levels were tested by biochemical. RT-PCR and Western blot method were used to detect nuclear factor PPAR-α, PG-C-1 mRNA and protein expression. Results: After Free fatty acid induced for 24h, oil red staining showed steatosis; TG content in model group were significantly higher than the control group (P〈0.01), no significant change in each group transaminase; and intracellular PPAR-α, PGC-lmRNA, protein expression decreased (P〈0.05, P〈0.01). Fresh onion extract treated groups compared with the control intraeellular TG was significantly lower (P〈0.01); PPAR- α, PGC-1mRNA and protein expression were significantly increased compared with the model group (P〈0.05). Conclusion: Onion extract of fresh had significant protective effects on cells of liver steatosis, which maybe related to PPAR- α, PCd2-1 gene up regulation.
