芎芍胶囊含药血清对人脐静脉内皮细胞中Ang-2及其受体Tie-2表达的影响

摘　　要：目的:探讨芎芍胶囊对人脐静脉内皮细胞(HUVEC)促血管生成素-2(Ang-2)及其受体Tie-2的分泌和mRNA表达的影响,以阐明其促进血管新生的可能机制。方法:芎芍胶囊高、中、低剂量含药血清处理HUVEC,倒置相差显微镜观察细胞生长状态;流式细胞术和MTT法观测HUVEC的增殖情况;ELISA法检测HUVEC分泌Ang-2和Tie-2的水平;RT-PCR法检测Ang-2及Tie-2的mRNA表达。结果:芎芍胶囊高、中、低剂量含药血清均有促进HUVEC增殖和Ang-2及Tie-2的表达,以及增加S期细胞比例作用。结论:芎芍胶囊含药血清能促进HUVEC的增殖及Ang-2、Tie-2的表达,可能是其促进血管新生的机制之一。Objective： To investigate the effect of Xiongshao Capsule（XSC） on angiopoietin-2 and its receptor Tie-2 secretion and mRNA expression of HUVEC,in order to clarify some of its angiogenic possible mechanism.Methods： HUVEC were incubated in vitro.XSC low,medium and high doses pharmaco-serum were identified to stimulated the proliferation of HUVEC and measured by inverted phase contrast microscope,MTT assay and FCM.To observe HUVEC secretion of angiopoietin-2（Ang-2）,Tie-2 by enzyme-linked immunosorbent assay（ELISA） method.To detected Ang-2,Tie2 expression of mRNA by RT-PCR.Results： The results showed XSC could promote HUVEC proliferation and increase the promotion of S phase cells.XSC could significantly increase Ang-2 and Tie-2 expression.Conclution： XSC could promote HUVEC proliferation and the expression of Ang-2,Tie-2.That may be its important role in angiogenesis one of the mechanisms.
