全蝎提取物对人前列腺癌PC-3细胞体外抑制作用研究

摘　　要：目的:研究全蝎提取物对人前列腺癌PC-3细胞的体外抑制作用。方法:以液氮快速冻融法及丙酮沉淀法制备全蝎蝎身、蝎尾水提物,不同浓度体外干预人前列腺癌PC-3细胞,四甲基偶氮唑盐比色法检测两种全蝎提取物对PC-3细胞增殖的影响并计算半数抑制浓度(IC50),流式细胞术检测提取物对PC-3细胞细胞周期的影响及凋亡诱导。结果:全蝎提取物以剂量依赖性方式抑制PC-3细胞增殖,其中蝎身提取物IC50为2.3 mg/mL,蝎尾提取物为0.8 mg/mL。全蝎提取物作用后的可诱导PC-3细胞凋亡,G0/G1期细胞比例下降,G2/M期、S期比例上升。其中蝎尾提取物作用更明显,相同药物浓度条件下诱导细胞凋亡作用与蝎身提取物比较,差异有统计学意义(P<0.01)。结论:全蝎提取物可抑制人前列腺癌PC-3细胞增殖及诱导凋亡,可使PC-3细胞停滞于S期,从而抑制生长。且蝎尾的作用强于蝎身,提示蝎毒可能是全蝎中发挥抗肿瘤作用的主要成分。Objective： To study the whole xie extract on people prostate cancer cells in vitro PC-3 inhibition.Methods： the method and the fast freeze-thaw liquid nitrogen acetone precipitation preparation whole xie scorpion body,scorpion tail water extractings from different concentration,in vitro intervention people prostate cancer cells,four-3 PC methyl accidentally nitrogen） salt colorimetric method detection two whole xie extract on PC-3 cell proliferation and calculate the influence of half inhibit concentration（IC50）,streaming cells during the extract on PC-3 detection cells of the cell cycle and apoptosis induced effects.Results： the whole xie extract to dose dependent way PC-3 inhibit cell proliferation,including body extract for 2.3 IC50 Capricorn mg/mL,scorpion tail extract for 0.8 mg/mL.Whole xie extract the role to induce PC-3 cell apoptosis,G0/G1 phase cells was down,G2/M period,S phase ratio rose.The role of scorpion extract more apparent,under the condition of the same drug concentration induce the apoptosis body function and scorpion extract comparison,difference have statistical significance（P0.01）.Conclusion： the scorpion can extract on prostate cancer PC-3 cell proliferation and apoptosis induction,can make the PC-3 cell stagnation in S phase,thus inhibiting growth.And scorpion tail role than the body,scorpion hints scorpid poison may be whole xie play antitumor function＇s main ingredient.
