摘　　要：为了研究西太平洋暖池核心区表层古菌群落结构及多样性,应用16S rDNA文库技术对KX08-973-22站位(取名为CWP)水体样构建基因文库,得到了561个克隆,经分析处理后得到50个OUT(OperationalTaxoni mic Units)。通过对16SrRNA基因序列的同源性比较,构建系统发育树分析及统计学分析,得出的结果:本次研究中的古菌类群都属于广古菌(Euryarchaeota)和泉古(Crenarchaeota)。广古菌以marine group II(MG II)(49.0%)为主,marine group III(MGIII)占一小部分(5.7%)。泉古菌以Marine Group I(MG I)为主(45.1%),Miscellaneous Crenarchaeotic Group(MCG)占极少数(0.2%)。本次研究的西太平洋暖池核心区表层古菌多样性不高,群落结构在小尺度上存在垂直变化。To study the archaeal community structure and diversity in the core area of West Pacific warm pool,the 16S rDNA library technology was employed to construct the clone library for the water samples from the KX08-973-22 site（named CWP）.A total of 561 clones were obtained and 50 operational taxonomic units（OTUs） were identified.By homologous comparison for the 16S rRNA gene sequences,construction of phylogenetic tree and statistical analysis,it has been found that all the archaeal phylotypes in this study are affiliated with Euryarchaeota and Crenarchaeota,respectively.Euryarchaeota is dominated by Marine Group II（MGII,49.0%）,with minor Marine Group III（MGIII） only accounting for 5.7% of the libraries.Crenarchaeota is dominated by Marine Group I（MGI）with tiny amount of the Miscellaneous Crenarchaeotic Group（MCG,0.2%）.Our study suggests that the archaeal phylotypes from the core area of the West Pacific warm pool are characterized with low diversity and community structure varies within small vertical distances.
