摘  要:目的:探讨解毒化瘀药含药血清对白血病K562/A02细胞mdr1耐药基因、核因子κB(Nuclear factor kappa B,NF-κB)信号基因表达的影响。方法:采用半定量逆转录聚合酶链反应法检测解毒化瘀药含药血清处理后K562/A02细胞mdr1基因的表达;Western blot法检测NF-κB/P65、NF-κB/P50、IκBα的表达。结果:解毒化瘀药含药血清能够降低K562/A02耐药细胞NF-κB/P65、NF-κB/P50、IκBα表达,降低mdr1耐药基因的表达。结论:解毒化瘀药对K562/A02细胞的耐药逆转作用可能是通过阻断NF-κB信号通路,影响mdr1耐药基因的表达起作用的。Objective:To discuss the influence of Jieduhuayu Chinese herbal serum on the expression of multidrug resistance gene1(Mdr1) of leukemia K562/A02 cell and the expression of NF-κB(nuclear factor kappa B).Methods:Semi-quantitative reverse transcriptase polymerase chain reaction method was used to examine the expression of Mdr1 gene after K562/A02 cell receiving the treatment of Jieduhuayu Chinese herbal serum;Western blotting technology was used to examine of the expressions of NF-κB/P65,NF-κB/P50,and IκBα.Results:The expressions of NF-κB/P65,NF-κB/P50,and IκBα and the expression of multidrug resistance gene1 of leukemia K562/A02 cell could be decreased by Jieduhuayu Chinese herbal serum.Conclusion:Jieduhuayu Chinese herbal serum can reverse multidrug resistance in leukemia cell by blocking NF-κB signal pathway,and influence the expression of multidrug resistance gene1(mdr1).
