摘  要:目的观察谷氨酰胺联合维生素C对肠源性内毒血症的预防作用,并初步探讨其机制。方法将160只SD雄性大鼠按随机数字表法分为谷氨酰胺联合维生素C组、谷氨酰胺组、维生素C组、高温应激组、正常对照组。用人工气候箱建立大鼠中暑内毒素血症模型,分别于1、3、7、15 d用2%的戊巴比妥钠腹腔麻醉取小肠组织并心脏采血。用试剂盒检测小肠组织超氧化物歧化酶(SOD)、丙二醛(MDA)及血清内毒素的含量,RT-PCR检测小肠黏膜Toll样受体2(TLR2)mRNA表达情况。结果在同时间段与正常对照组比,高温应激组SOD含量明显降低,MDA、血清内毒素、TLR2mRNA表达量明显升高(P<0.05),3种治疗组SOD、MDA含量无统计学差异(P>0.05),谷氨酰胺组、维生素C组血清内毒素及TLR2 mRNA表达量明显升高(P<0.05),但谷氨酰胺组联合维生素C组血清内毒素及TLR2 mRNA表达量无统计学差异(P>0.05)。在同时间段与高温应激组相比,3种治疗组SOD含量显著升高(除第7天谷氨酰胺组、维生素C组外);MDA、内毒素含量及TLR2 mRNA表达量明显降低(除第3天、第15天的维生素C组TLR2 mRNA表达量外)(P<0.05)。同时间段的谷氨酰胺联合维生素C组与谷氨酰胺组、维生素C组相比,其SOD含量相近,但MDA和血清内毒素含量及TLR2 mRNA表达量均显著降低(P<0.05)。结论谷氨酰胺联合维生素C可减轻高热应激对肠黏膜的损伤,保护肠黏膜屏障功能,有效的降低肠源性内毒素血症发生,可用于高热中暑的预防。Objective To observe the preventive effect of pretreatment of glutamine combined with vitamin C on intestinal endotoxemia,and to investigate the mechanism in heat stroke prevention.Methods One-hundred-and-sixty male SD rats were randomly divided into a glutamine and vitamin C group,a glutamine group,a vitamin C group,a heat stress group,and a normal control group.A rat model of heat stroke-induced endotoxemia was built using an artificial climate chamber.At 1,3,7 and 15 d after heat stroke,the small intestine tissues and heart blood of the rats were collected after intraperitoneal anesthesia with 2% sodium pentobarbital.The levels of superoxide dismutase(SOD) and malondialdehyde(MDA) in small intestine tissues and serum endotoxin level were measured using test kits.The mRNA expression of Toll-like receptor 2(TLR2) in small intestine mucosa was tested by RT-PCR.Results Compared with the normal control group,SOD level decreased,while MDA level,serum endotoxin level,and TLR2 mRNA expression increased significantly in the heat stress group(P0.05).There were no statistical differences in SOD level and MDA level between the three treatment groups and the normal control group.Compared with the normal control group,serum endotoxin level and TLR2 mRNA expression increased significantly in the glutamine group and vitamin C group(P0.05),but had no statistical differences in the glutamine and vitamin C group.Compared with the heat stress group,SOD level increased significantly in the three treatment groups(except for the glutamine group and vitamin C group at 7 d after heat stress),while MDA level,serum endotoxin level,and TLR2 mRNA expre-ssion decreased significantly(except for TLR2 mRNA expression in the vitamin C group at 3 d and 15 d after heat stress)(P0.05).Compared with the glutamine group and vitamin C group,SOD level was similar in the glutamine and vitamin C group,but MDA level,serum endotoxin level,and TLR2 mRNA expression decreased significantly(P0.05).Conclusion Glutamine comb
