摘 要:目的:观察丹红通精方对自身免疫性慢性前列腺炎大鼠血液及前列腺组织中细胞因子水平的影响。方法:选取8~10周龄Wistar雄性大鼠40只,随机分成空白对照组(n=10)、低剂量实验组(n=10)、中剂量实验组(n=10)及高剂量实验组(n=10),其中,实验组予大鼠前列腺蛋白提纯液诱导造模。造模成功后,空白对照组予生理盐水灌胃,各实验组分别给予低、中、高剂量丹红通精方混悬液灌胃处理,疗程35 d。取大鼠颈动脉血及前列腺组织。采用双抗体夹心酶联接免疫吸附剂测定(ELISA)法定量分析各组大鼠血清中肿瘤坏死因子-α(TNF-α)、白介素-10(IL-10)的表达,实时荧光定量RT-PCR检测各组大鼠前列腺组织中TNF-α、IL-10 mRNA的表达。结果:干预后,空白对照组与各实验组(低、中、高剂量)大鼠血清中TNF-α、IL-10含量分别为(20.18±9.12)、(86.78±30.25)ng/L,(23.89±8.07)、(94.99±35.02)ng/L,(111.46±6.71)、(130.17±43.61)ng/L,(138.52±5.75)、(195.22±56.34)ng/L。四组大鼠前列腺组织中TNF-α、IL-10 mRNA含量分别为(0.17±0.02)、(0.58±0.11)ng/L,(0.27±0.02)、(0.81±0.03)ng/L,(0.35±0.11)、(0.93±0.19)ng/L,(0.98±0.03)、(0.99±0.34)ng/L,各组比较,差异均有统计学意义,其中,中、高剂量实验组差异有高度统计学意义(P<0.05或P<0.01)。结论:丹红通精方能提高前列腺蛋白提纯液诱导的自身免疫性慢性前列腺炎大鼠血清及前列腺组织中TNF-α、IL-10水平,并且与低剂量丹红通精方相比,中、高剂量丹红通精方的作用更加明显。Objective: To observe the influence of Danhongtongjing prescription on cytokines levels of blood and prostate tissue in autoimmune chronic prostatitis rats. Methods: 40 male Wistar rats which were 8-10 weeks old were randomly divided into blank control group (n=10), low dose group experiment group (n=10), medium dose experiment group (n=10) and high dose experiment group (n=10). The rat models were induced by using the prostate protein purified. The blank control group was given saline by intragastric administration, and the experiment groups were given low, medium and high dose of Danhongtongjing prescription suspension by intragastric administration, respectively. The course of treatment was 35 days. The carotid artery and prostate tissue of rats were collected. The expression of tumor necrosis factor-α (TNF-α) and interleukin -10 (IL-10) in rats serum were quantitative analysed by double antibody sandwich enzyme immunosorhent assay (ELISA).The expression of TNF-α and [L-10 mRNA in rat prostate tissue were detected by real-time fluorescence quantitative PCR. Results: After the intervention, the TNF-α and IL-10 contents in rats serum of the control group and the each experiment group (low, medium and high dose) were (20.18±9.12) and (86.78±30.25)ng/L, (23.89±8.07) and (94.99±35.02)ng/L, (111.46±6.71) and (130.17±43.61)ng/L, (138.52±5.75) and (195.22±56.34)ng/L, respectively. The TNF-α, IL- 10 contents in prostate tissue of the control group and the each experiment group were (0.17±0.02) and (0.58±0.11)ng/L, (0.27±0.02) and (0.81 ±0.03)ng/L, (0.35±0.11) and (0.93 ±0.19)ng/L, (0.98±0.03) and (0.99±0.34)ng/L, respectively. The differences had statistical significance, especially the differences in medium dose experiment group and high dose experiment group (P〈0.05 or P〈0.01).
