补体C3在男性不育患者血浆蛋白中的差异分布

摘　　要：目的:分析男性不育患者与正常捐精者血浆蛋白含量的差异。方法:利用低浓度的聚丙烯酰胺凝胶(SDS-PAGE)电泳分离10例正常捐精者和2例不育症男性患者的血浆蛋白,对差异蛋白条带进行基质辅助激光解析电离飞行时间质谱(MALDI-TOF/TOF-MS)鉴定,继而对142例不育男性患者血浆进行分析。结果:男性不育患者与正常捐精者血浆蛋白SDS-PAGE电泳结果显示,在分子量约为200kD及120kD处出现了两条明显的蛋白差异条带,经鉴定为补体C3。对所有144例不育患者的血浆蛋白分离验证结果表明,同时出现这两条差异条带的有135例(93.75%)。结论:补体C3在男性不育患者与正常捐精者血浆中存在蛋白形式差异,该差异在不育患者中的生物学意义有待进一步研究。Objective ： To investigate different expressions of plasma proteins between male infertility patients and normal fertile men. Methods： Sodium dodecyl sulphate polyacrylamide gel electrophoresis （SDS- PAGE） was used to separate plasma proteins of 2 male infertility patients and 10 normal donors, followed by identification through matrix - assisted laser desorption/ionization time of flight （MALDI -TOF） mass spectrometry. Plasma samples of 142 male infertility patients were evalua- ted. Results： The SDS -PAGE analysis showed two different bands locating at 200 kD and 120 kD, which were identified as complement component 3. Among 144 samples of male infertility patients, 135 （93.75%） were found these two identified bands. Conclusion： The difference of plasma complement component 3 between male infertility patients and normal fertile men would be used to evaluate male infertility.
