摘  要:目的:探讨天花粉蛋白(TCS)对卵巢癌细胞株HO8910增殖效应并探讨其机制。方法:采用MTT法检测不同浓度(20、40、80μmol/L)对细胞HO8910增殖的影响;通过流式细胞仪观察细胞(20μmol/L)凋亡过程中细胞周期的变化;Western Blot检测不同浓度(20、40、80μmol/L)72hTCS对HO8910凋亡相关蛋白Bax、Bcl-2的表达。结果:随着TCS浓度的升高,作用时间的延长,TCS对HO8910生长抑制率逐渐增高(P<0.05),呈一定的时间,剂量依赖性。流式细胞仪检测随着天花粉蛋白浓度的增加,Bax基因的表达增加而Bcl-2的表达减少。结论:TCS对卵巢癌细胞HO8910细胞增殖抑制作用,该作用可能与其下调Bcl-2及上调Bax蛋白表达有关。Objective :To investigate the effect of trichosanthin on ovarian cancer cell line HO8910 inhibiting the proliferation effect and apoptosis - inducing effects and explore its apoptosis - related genes Bax and Bcl - 2 expression. Methods : MTT assay of drugs on cells in vitro ;flow cytometry cell apoptosis in the cell cycle changes. Western Blot Detection of apoptosis -related proteins Bax, Bcl - 2 expression. Results : With the drug concentration increased, the role of the time extension of triehosanthin on HO8910 cell growth inhibition rate was gradually increased ;under the effect of triehosanthin, HO8910 cells apoptotic morphological changed and apoptotic cells histological changed. Flow cytometry histogram also appears characteristic of the sub - diploid peak, with the increase in the concentration of trichosanthin, Bax gene expression increases the expression of Bcl - 2 reduction. Conclusion:Trichosanthin can induce HO8910 ovarian cancer cell apoptosis. Bcl -2 protein in the reduction of Bax protein expression increasing apoptosis in ovarian cancer cells may be one of the mechanisms.
