安胃二号方对胃溃疡大鼠BCL-2及NF-κB的影响

摘　　要：目的:研究安胃二号方对胃溃疡的影响,并探讨其机制。方法:40只wistar大鼠,随机分为空白对照组、病理模型组、奥美拉唑组、安胃二号方组。采用改良Takagi法制作乙酸胃溃疡大鼠模型,治疗13天后测量各组溃疡指数,用免疫组化法检测胃黏膜细胞Bcl-2、NF-κB蛋白的表达。结果:安胃二号方组与其他组比较溃疡指数明显降低(P<0.01),安胃二号方组与奥美拉唑组Bcl-2的表达明显高于模型组(P<0.01),且安胃二号方组高于奥美拉唑组(P<0.05)。安胃二号方组NF-κB的表达明显低于模型组(P<0.01)。结论:安胃二号方能通过增加Bcl-2蛋白表达,抑制NF-κB蛋白的表达,起到对胃溃疡的保护作用,这可能是其促进溃疡愈合机制之一。Objective ： To study the protective effects of Anwei Decoction - 2 （ AD - 2） on gastric ulcer of rats and its effective mechanism. Methods ： Forty healthy wistar rats were randomly divided into model group, AD - 2 treatment group, omeprazole treatment group and healthy group. The rat model of gastric ulcer was set up by Takagi＇ s method with some modification. After treating 13d,surveys each group of ulcer index,and the expression of Bcl -2 and NF - KB in the gastric ulcer tissues was examined by immunohistochemical method. Results：AD -2 treatment group could obviously decrease the ulcer index than the others group （P 〈 0.01 ）. Cmpared with the model group, the expression of Bcl -2 was evidently enhanced in the AD -2 treatment group and aomeilazuo treatment group （P 〈 0.01 ）. Significant difference was also noted in the expression level of Bcl -2 between the two treat- ment groups（ P 〈 0.05 ）. Cmpared with the model group, the expression of NF-κB was evidently decrease in the AD -2 treatment group （P 〈 0.01 ）. Conclusion：The results indicated that AD -2 has significantly protection on gastric ulcer. The one of mechanism of its promoting the healing of the gastric ulce may be that the AD - 2 can increase the expression of Bcl - 2 and reduce NF -κB expression.
