尼可地尔对动物心血管的影响

摘　　要：尼可地尔静脉注射50μg/kg,能增加犬的冠脉血流量和降低冠脉阻力;家兔静脉注射尼可地尔100μg/kg,能明显解聚血小板的聚集。恒速滴入法灌注0.1-1.0mmol/L尼可地尔,使豚鼠心肌动作电位复极时程(AFD_(50)、APD_(90))有不同程度的缩短,有效不应期(ERP)亦明显缩短。适当调整剂量(200μg/kg)对犬可降低心肌氧耗和左心室负荷,从而心肌收缩力得到改善,提示尼可地尔可作为治疗冠心病药物。In dogs, nicorandil 50 ug/kg iv increased the coronary blood flow and reduced its vascular resistance.In rabbits, nicorandil 100 ug/kg iv decreased the aggregation of the platelet.In guinea pigs, APD (action potential duration)80, APD,ERP(effective refractory period) were all reduced by per fusion of isolated myocardial strip with nicorandil, adjusting to proper dose(200 ug/kg)in dogs reduced the myocardial oxygen demand and the left ventricular load, so that the myocardial contractility was improved.Nicorandil is recommended to treat coronary disease.
