摘 要:目的建立酒精性肝病大鼠血浆中PDGF含量的HPLC测定方法。方法色谱柱为Hypersil C18柱(250 mm×4.6 mm,5μm),流动相为乙腈-水,梯度洗脱(5%~ 65%,40 min),流速1.0mL/min,柱温30℃,检测波长210 nm,进样量10μL。结果 PDGF线性范围浓度在0.01~0.30μg/mL时与峰面积呈良好的线性关系(r=0.999 3),测定方法回收率78.6%,RSD<15%。结论 该方法简便、快捷,可用于酒精性肝病大鼠血浆中PDGF的含量测定。Objective To establish an HPLC method for determination of platelet-derived growth factor (PDGF) level in rats plasma. Methods The HPLC analysis was performed on the Hypersil C18 column (250 mm ×4.6 mm, 5 μm) at temperature 30℃.The mobile phase consisted of acetonitrile and water, using gradient elution (5% - 65%, 40 min) with the flow rate 1.0 mL·min^-1. The detection wavelength was 210 nm. Results The standard curve was linear in the range of 0.01-0.30μg·mL^-1 (r =0.999 3). The method recovery was 78.6%, and the intra-day and inter-day precisions were less than 15%. Conclusion The method is simple,convenient and rapid, which could be used to determine the PDGF in alcoholic hepatitis rats plasma.
