摘  要:目的研究利用大孔吸附树脂富集纯化野马追总黄酮的工艺条件及参数。方法以总黄酮吸附量、醇洗物总黄酮含量、总黄酮回收率为考察指标,考察D101大孔吸附树脂分离纯化野马追总黄酮的动态吸附洗脱条件。结果野马追提取物上样总黄酮浓度为6 mg/mL,pH调节为5~ 6时,动态吸附容量达65 mg/g(11.7 mg/mL),吸附流速以1 BV/h为宜,6倍量柱体积70%乙醇为洗脱剂,以5 mL/min的(相当于3BV/h)流速进行洗脱,树脂可重复使用5次为最佳次数。结论 本方法可方便有效地纯化野马追总黄酮,所得野马追总黄酮得率2.71%,含量72.13%。Objective To optimize and establish the technology purifying total flavonoids from Eupatorium lindleyanum produced in Jangsu with macroporous absorption resin D101. Methods With the yield of total flavonoids as index ,to screen refining process of extracting from adsorbent of D101 macroporous resin by eluting with ethanol of different concentrations, setting content of flovonoids as parameter. Results The D101 macroporous resin had the best separation efficiency when the total flavonoids in the sample concentration was 6mg/mL.pH was 5-6, D101 macroporous resin adsorption flow 1BV/h dynamic absorption capacity was determined to be 65 mg·g^-1 (11.7 mg/mL),eluting with 6BV volume of 70% ethanol as desorptive and desorption flow 3BV/h. Conclusion The yield and the content of total flavonoids from Eupatorium lindleyanum produced in Jangsu were 2.71% and 72.13%, respectively. This method is advisable.
