摘 要:目的探讨姜黄素衍生物FM0817体外抗多药耐药鲍氏不动杆菌作用及其机制。方法应用VITEK-2Compact系统检测18株多药耐药鲍氏不动杆菌对抗菌药物的敏感性、β-内酰胺酶类和氨基糖苷类药物耐药表型,PCR扩增相应耐药基因,采用滤纸片扩散法初步判断FM0817是否有抗菌作用,作用结果结合耐药表型和基因型,分析FM0817体外抗多药耐药鲍氏不动杆菌作用的机制。结果鲍氏不动杆菌多药耐药现象明显,FM0817抗多药耐药鲍氏不动杆菌作用明显。结论姜黄素衍生物FM0817具有较强的体外抗多药耐药鲍氏不动杆菌作用,有进一步研究开发的前景;其作用机制与多药耐药性鲍氏不动杆菌耐药表型和基因型未发现有关系,可能存在新的作用靶点。OBJECTIVE To analyze the antibacterium activity of curcumin derivative FM0817 and its mechanism to Acinetobacter baumannii(ABA).METHODS A total of 18 strains of ABA were tested their susceptibilities to antibiotics by dilution methods.Total DNA was extracted by boiling methods.The aminoglycoside and β-lactamases drug resistance phenotypes in 18 strains MDR-ABA were detected by senior expert system of VITEK 2 Compact.18 ABA strains were used as a template in PCR to amplify the resistance genes of β-lactamases and aminoglycoside modified enzymes.RESULTS The susceptibilities tests showed that the multi-drug resistance of ABA was obvious.FM0817 exhibited potent antibacterial activity in vitro to 18 strains of multi-drug resistant ABA.CONCLUSION FM0817 exhibits potenter antibacterial activity in vitro,and it deserves further research and development,We have not found the relationship among mechanism of FM0817 and the multi-drug resistance phenotype and the resistance genes phenotype of A.baumannii.
