摘  要:目的观察复方丹参片对急性脑缺血大鼠的治疗效果,探讨其对急性脑缺血大鼠的保护作用和机制。方法采用Longa改良栓线法制备大鼠大脑中动脉缺血模型。实验大鼠随机分为正常组、假手术组、模型组、血塞通胶囊组、复方丹参片组5组,术后后正常组、假手术组、模型组予蒸馏水灌胃。血塞通胶囊组予血塞通胶囊灌胃,复方丹参片组予复方丹参片灌胃。分别在24h,3d,7d等时间点对以上5组进行神经功能缺损评分。断头取脑,TTC染色观察脑梗死面积,免疫组织化学方法及积分光密度值观察梗死周围区脑组织血管新生因子蛋白的表达。结果复方丹参片可明显改善急性脑缺血大鼠的行为学评分,减少梗死面积百分率,显著增加大鼠脑内血管新生因子蛋白的表达,促进急性脑缺血大鼠脑缺血组织侧支循环的建立。结论复方丹参片增强急性脑缺血大鼠梗死灶周围血管生长因子的表达,促进了梗死灶周围的血管新生。Objective To observe the therapeutic effect of compound Danshen tablet on acute cerebral infarction and explore its mechanisms.Methods The middle cerebral artery occlusion(MCAO) rat models were established by reforming Longa method.Rats were divided randomly into normal group,ischemia model group,sham operation group administered intragastrically with distilled water,ischemia model with Xuesaitong tablet treatment group administered intragastrically with Xuesaitong tablet,and compound Danshen tablet treatment group administered intragastrically with compound Danshen tablet.The cerebral area and neurological deficit were determined by triphenyl tetrazolium chloride(TTC) staining and Longa S score.The vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(bFGF) protein were detected by immunohistochemistry and in situ hybridization.Results Compound Danshen tablet had protective effects for cerebral ischemia through upregulating the expression of VEGF and bFGF.It promoted collateral circulation in ischemic tissue of acute cerebral ischemia.Conclusion Compound Danshen tablet could increase the expression of endothelial growth factor and promote the angiogenesis in rats with cerebral ischemia.
