摘 要:目的探讨电离辐射诱导人外周血GADD45A基因的表达变化,为寻找急性照射条件下辐射损伤早期诊断标志物提供实验依据。方法采集无急慢性疾病、不抽烟、年龄在26~29周岁的3名男性非放射性工作者外周血各10ml,分为10组,每组1ml。利用中国辐射防护研究院60Co照射装置对采集的血样分组照射,剂量率为0.313Gy/min。吸收剂量分别为0.05、0.1、0.2、0.4、0.6、0.8、1、2和3 Gy,照射后的人外周血于37℃培养箱中静置2h后提取RNA,利用实时荧光定量PCR技术对外周血GADD45A基因进行检测。结果实时荧光定量PCR检测结果表明各剂量组与对照组相比,其基因表达均有显著变化。0.05~0.6Gy剂量组与对照组相比基因表达下调,其中0.05~0.2Gy之间,随着照射剂量的增加,基因表达量逐渐降低;0.2~0.6Gy之间,随着照射剂量的增加基因表达量又逐渐增加。0.8Gy时,基因表达量增幅较大,高于对照组和1Gy剂量组。1~3Gy剂量组基因表达量均高于对照组,且随着剂量增大,表达量增加,具有明显剂量相关性。结论电离辐射可以诱导人外周血GADD45A基因的差异表达,1~3Gy之间具有明显剂量相关性。0.05~0.2Gy之间外周血淋巴细胞表现出辐射刺激效应。Objective To explore the changes of GADD45A gene expression in human peripheral blood induced by ^60Coγ ray irradiation and offer the fundamental materials for the research of boimarker in early diagnosis of radiation damage. Methods Human peripheral blood was irradiated to 0.05 - 3Gy ^60Coγ - ray and changes of GADD45A genes in peripheral blood were detected by Real - time fluorescent quantitative PCR. Results After irradiation, the expression of GADD45 A genes in human peripheral blood changes significantly. Specifically, from 0. 05Gy to 0.6Gy, the expression of GADD45A genes decreased with the increase of irradiation dose. And from 1 Gy to 3Gy, the expression of GADIM5A genes increased with the increase of irradiation dose compared with the control. Conclusion 60 ^60Coγ ray irradiation can induce the changes of GADIM5A gene expression in human peripheral blood. And the changes of GADD45A gene shows fine dose - effect relationship from 1Gy to 3Gy. From 0. 05Gy to 0.2 Gy, peripheral blood lymphoeytes showed stimulation effect.
