摘 要:[目的]筛选分离纯化黄芪总皂苷的最佳工艺。[方法]利用动态吸附-解析方法,以紫外分光光度法测定黄芪总皂苷的含量,研究最适大孔树脂分离纯化黄芪总皂苷的条件。[结果]AB-8型大孔树脂富集纯化黄芪总皂苷的最优条件为:采用黄芪提取液浓度为0.1g/ml上柱,上样量为3 BV,流速为1.0 ml/min,解吸时先用4 BV蒸馏水淋洗,再用4 BV50%乙醇解吸,收集50%乙醇洗脱液,得黄芪总皂苷提取物浸膏,黄芪皂苷收率可达52.1%。[结论]该工艺简单可行,能满足大生产的要求。[Objective]To screen the optimum conditions for separation and purification of total saponins from RADIX ASTRAGALI.[Method]The content of total saponins in RADIX ASTRAGALI was determined by ultraviolet spectrophotometry using dynamic adsorption-desorption method aimed at studying the optimum conditions for separating and purifying total saponins from RADIX ASTRAGALI with macroporous resin.[Result]The optimum conditions for separation and purification of total saponins with AB-8 were as follows: extracting concentration of 0.1 g/ml,sampling volume of 3 BV,flow rate of 1.0 ml/min,washing with 4 BV of distilled water first and then desorption with 4 BV of 50% ethanol.Under the above condition,the yield of total saponins reached as high as 52.1%.[Conclusion]This method was simple and feasible,which could meet industrial requirements.
