HLA-DQA1基因与新疆吐鲁番地区维吾尔族2型糖尿病关联性研究

摘　　要：[目的]探讨新疆吐鲁番地区维吾尔族2型糖尿病与HLA-DQA1等位基因的关联性。[方法]采用聚合酶链反应-序列特异性引物技术,对110例新疆吐鲁番地区维吾尔族2型糖尿病患者和同地区107名维吾尔族正常对照者的HLA-DQA1等位基因频率进行检测,分析2型糖尿病HLA-DQA1等位基因多态性,并将所得结果与国内其他民族的同类资料进行比较。[结果]新疆吐鲁番地区维吾尔族2型糖尿病组与维吾尔族正常对照组比较,HLA-DQA1*0104(P=0.002,RR=9.802)等位基因频率明显增高,差异有统计学意义。新疆吐鲁番地区维吾尔族2型糖尿病患者与国内其他民族的同类资料进行比较,等位基因分布频率上有共同点,也有一定的独特性。[结论]HLA-DQA1*0104可能是新疆吐鲁番地区维吾尔族2型糖尿病的致病基因;新疆吐鲁番地区维吾尔族2型糖尿病患者与国内其他民族群体的资料比较,总体检验差异有统计学意义(P﹤0.01),显示人类不同民族的遗传易感性不一样,表现出民族之间的差异。[Objective] To investigate the association between the polymorphism of HLA-DQA1 alleles and type 2 diabetes in Uygurs nationality of Turpan in Xinjiang province. [ Methods] Polymerase chain reaction with sequence--specifiC primers （PCR-SSP） genotyping method was conducted in 110 ethnic Uygurs patients with type 2 diabetes mellitus and 10＂7 ethnically matched controls from Turpan of Xinjiang. Then a study was made on the association between the polymorphism of HLA-DQA1 alleles and type 2 diabetes mellitus and compare Uygurs of Turpan in Xinjiang with other nationalities from China. [ Results] The frenquency of HLA-DQAI＊0104 allele in the patients with type 2 diabetes mellitus were significantly higher than that in the healthy control. The frequencies of HLA-I）QA1 alleles in Uygurs of Turpan in Xinjiang were different with ether nationalities from china. [Conclusion] In Uygnrs nationality of Turpan in Xinjiang, HLA-DQAI＊0104 allele may be susceptible genes. Compared with Guangxi Zhuang and Yunnan Yi nationalities, the results suggested that there were common characterizations and individual differences in susceptible genes of different nationalities of human being.
