摘  要:目的:研究Musashi-1基因(MSI-1)启动子区-2297T>C(rs3742038)多态与我国人群食管癌发病的关联。方法:应用病例对照研究方法,采用Taqman技术检测300对食管癌病例的基因与正常对照-2297 T>C多态位点的基因型,并用SAS9.13软件分析该位点变异与食管癌易感性的关联。结果:以-2297TT基因型携带者为参照,携带C变异基因型(-2297CT/CC)个体发生食管癌的风险将显著增加(adjusted OR=2.27;95%CI=1.29-4.02),且随着变异等位基因个数增加,食管癌发病风险呈剂量—效应关系地增加(P_(trend)=0.0088)。结论:MSI-1基因启动子区-2297T>C基因变异型能显著增加我国人群发生食管癌的风险性,将为食管癌高危人群的筛选提供重要的遗传标志。Objective:To investigate the correlation between Musashi-1 ( MSI-1 ) gene promoter -2297T 〉 C (rs3742038) polymorphism and risk of esophageal carcinoma in Southern Chinese population. Methods:In a case-control study, the genotypes of-2297 T 〉 C polymorphism were studied by Taqman among 300 patients with esophageal carcinoma and 300 age- and sex frequency-matched cancer-free controls. SAS 9. 13 was used to analyze the correlation between this polymorphism and risk of esophageal carcinoma. Results : Compared with the- 2297 TY genotype carriers, the variant genotypes -2297CT/CC carriers had an increased risk of esophageal carcinoma( adjusted OR = 2.29 ;95% CI = 1.29-4. 18 ). The risk of esophageal carcinoma increased along with the number of variant alleles in a dose-effect relationship( Ptrend = 0.0008 ). Conclusion:The variant genotypes (-2297CT/CC) in promoter region of MSI-1 gene are associated with an increased risk of esophageal carcinoma in Southern Chinese population, which can be used as a biomarker in screening of high risk population for esophageal carcinoma.
