摘  要:目的观察三碘甲腺原氨酸(T3)对氟他胺(Flu)诱导的SD大鼠隐睾生殖母细胞(Go)发育及神经细胞黏附分子(NCAM)表达的调控作用。方法 15μg/(kg.d)T3皮下注射Flu诱导出生后第1 d(PD1)仔鼠、PD20隐睾仔鼠,观察干预后隐睾发生率、组织学改变,免疫组化及逆转录聚合酶链反应(RT-PCR)检测NCAM表达。结果 Flu诱导仔鼠PD1给予T3干预,成年后隐睾发生率为25%(8/32),较未干预组的隐睾发生率40.9%(9/22)降低,且隐睾曲细精管内无残留生殖母细胞(Go),NCAM阴性表达。Flu诱导的PD13仔鼠睾丸组织、PD20仔鼠隐睾组织NCAMmRNA表达高于正常组(P<0.01,P<0.05),T3干预后,NCAMmRNA表达均低于非T3干预组(P<0.01),并与正常组无明显差异(P>0.05)。结论 T3可以降低Flu诱导暴露后隐睾发生率,下调NCAM表达,降低或消除隐睾曲细精管Go残存。Objective To investigate the regulation of T3 on the maldeveloped gonocytes(Go) and the expression of NCAM in Flutamide-induced cryptorchidism SD rat.Methods Postnatal day(PD) 1 and PD20 cryptorchidism SD rat induced by Flu were treated with 15 μg/(kg·d) T3 by subcutaneous injections.Incidence of the cryptorchidism,histology were evaluated.And the expression of NCAM was detected by immunohistochemisty and RT-PCR.Results The incidence of cryptorchidism in the group of Flu-induced offsprings treated by 15 μg/(kg·d) T3 from PD1 was 25%(8/32),which was lower than that of Flu group 40.9%(9/22).No Go and NCAM expression were observed in the seminiferous tubules.NCAM mRNA expression in Flu-induced PD13,PD20 testes was up-regulated when compared with control(P0.01,P0.05),while after treated with 15 μg/(kg·d) T3 the NCAM expression was significantly down-regulated compared to that of non T3 treated group(P0.01),and was not significantly different from that of control(P0.05).Conclusion The incidence of Flu-induced cryptorchidism can be decreased by T3 treatment after birth.T3 may contribute to the down-regulation of the NCAM expression,and reduction or elimination of the remainded gonocytes in crytorcidism seminiferous tubules.
