摘  要:目的:建立近红外光谱快速测定龙胆药材功效成分的方法。方法:在10 000~4 000 cm-1光谱范围内扫描样品,以交叉验证均方根误差(RMSECV)和决定系数(R2)为指标,通过筛选,确定了用于建模的最优近红外波段和光谱预处理方法。采用偏最小二乘法建立NIR与HPLC信息间的关系。结果:龙胆苦苷与獐牙菜苦苷近红外预测值与真实值的R2分别为0.9534,0.8135,RMSECV为0.184,0.0177,裂环烯醚萜苷总量的决定系数和校正均方差为0.9435,0.211。结论:该模型可直接对样品进行快速准确的检测,具有非破坏性、无污染等优点,可以应用于龙胆药材的定量测定。Objective: The quantitative analysis model for determination of the content of Radix Gentiana scabra Bge. was established using near-infrared diffuse reflectance spectroscopy. Methods: NIR spectra of the samples were collected in range of 10 000 to 4 000 -1 cm Different wave number ranges and spectrum preprocessing methods were investigated according to the root mean square errors of correction values (RMSECV) and the correlation coefficient (R2). Multivariate calibration models based on PLS algorithm were devel- oped to correlate the spectra and corresponding values determined by HPLC. Results: The R2 between true and prediction were 0. 9534,0. 813 5,0. 943 5. The RMSECV were 0. 184, 0. 017 7, 0. 211, respectively. Conclusion: It can be used rapidly and correct- ly for determination of Radix Gentiana scabra Bge. It owns many remarkable advantages that can not be displayed by traditional analysis methods, such as nondestruction, no pollution. NIRDS can control the quality of Radix Gentiana scabra Bge.
